1475318 Testing blood BLOCK ENG Inc 19 April 1974 [31 May 1973 30 Jan 1974] 41066/76 Divided out of 1475317 Heading G1A A system for differentially counting leukocyte types randomly distributed in a blood medium comprises (a) means (not shown), for staining the leukocyte types with a dye composition comprising a plurality of dyes, at least one of the dyes imparting a first fluorescence to the nuclei of all leukocytes and another dye imparting a second fluorescence to some leukocyte types, (b) flow means 52 through which the stained leukocytes in the blood medium under test are to pass in single file, (c) source means 54 &c. for illuminating the stained leukocytes passing through the flow means, with radiation in the characteristic absorption wavelength region of each of the dyes, and (d) detection means 81, 74, 80 optically coupled to the flow means for detecting the leukocytes passing through the flow means in single file, the detector means distinguishing and counting the leukocyte types according to the relationships between the intensities of the radiation emitted by each leukocyte type in characteristic emission wavelength regions. In the embodiment shown, source 54 is a mercury arc lamp, and the radiation passes through collimating lens 56, reflecting filter 58, for example a dichroic filter, condensing lens 64 or 72 to focus on capillary tube flow path 52 at 62 or 70. Photo-multipliers 74, 80 and photo-diode 81 detect emitted radiation in different respective wavelength ranges and the signals from the photo-electric devices are passed to processor 86 where in the embodiment described there is calculated the ratio of the measured intensities of radiation of green and of red wavelengths to distinguish the different leukocyte types for counting. Display device 88 shows the results. In a related embodiment, Fig. 1 (not shown), the leukocytes are stained in the form of an alcoholfixed blood smear on a slide and the incident and transmitted radiation paths each include a filter wheel (18, 28), which carries two filters capable of transmitting light in different wavelength bands. The detection system comprises an image scanning device. The invention may be used to distinguish and count erythrocytes, reticulocytes and leukocyte types in a blood sample.
