1460028 Producing 3<SP>1</SP>,5<SP>1</SP>-cyclic adenylic acid by fermentation KIKKOMAN SHOYU CO Ltd 15 Oct 1973 [16 Oct 1972 23 Feb 1973] 48515/73 Heading C2P 3<SP>1</SP>,5<SP>1</SP>-Cyclic adenylic acid (CAMP) is obtained by culturing under aerobic conditions a mutant strain of a microorganism in a nutrient medium containing a source of carbon, a source of nitrogen and one or more inorganic salts, and recovering the 3<SP>1</SP>,5<SP>1</SP>-cyclic adenylic acid accumulated in the nutrient medium, the mutant strain being one obtainable by subjecting a microorganism capable of producing CAMP in the presence of a precursor and/or of a fluoride to mutation treatment and being one which is capable of producing CAMP in substantial amount in the absence of a precursor, a fluoride or a fluoride and a precursor. Specified parent strains from which suitable mutants can be obtained are Corynebacterium murisepticum No. 7 (ATCC 21374), Arthrobacter 11 (ATCC 21375) and Microbacterium No. 205 (ATCC 21376). The mutant strains may be obtained from the parent strain by conventional artificial mutation treatment, e.g. by subjecting the parent strain to X-radiation, irradiation with U.V. light or to treatment with a chemical mutagenic agent, e.g. nitrosoguanidine, diethyl sulphate, methylethyl sulphate, NaNO 2 , acridine or nitrogen mustard. The nutrient medium may contain a precursor and/or fluoride or may be free of both a precursor and fluoride. Several precursors which may be added to the nutrient medium are specified and the medium may also contain one or more cyclic 3,5<SP>1</SP>-nucleotide phosphodiesterase inhibitors several of which are specified. The culturing is preferably carried out at a pH of from 5-9 and at 20‹ to 40‹ C. The CAMP can be isolated by treatment with active carbon or with a cationic or anionic exchange resin or by addition of a solvent in which CAMP is insoluble or by a combination of two or more such treatments. Examples are given for preparing suitable mutants and their use in the production of CAMP.
