Abstract
1414640 Purification of #-amylase AJINOMOTO CO Inc 22 Dec 1972 [25 Dec 1971] 59475/72 Headings C3H and C3U Microbial #-amylase is recovered from aqueous solutions thereof by contacting an aqueous microbial #-amylase solution with starch until the #-amylase is adsorbed on said starch and thereafter recovering the #-amylase adsorbed on the starch. The preferred #-amylase is that produced by the genus Bacillus, e.g. Bacillus polymyxa ATCC 8523, Bacillus megaterium FERM P-1201 or FERM P-1202. The enzyme is preferably contacted with the starch in the presence of 5-15% by wt. per volume of ammonium sulphate and may be removed from the starch by suspending the starch/enzyme adduct in water, removing the starch and recovering the pure #-amylase. Starch may be saccharified by treatment with the #-amylase adsorbed on starch wherein prior to use the #-amylase adsorbed on starch is warmed in a dilute solution of ammonium sulphate. A method for the saccharification of starch comprises adding to a starch to be saccharified a #-amylase obtained as above and which has been adsorbed on starch, warmed to 40-60‹ C. in an ammonium sulphate solution having a concentration of 0À5-2% and then removing the sugar produced.
