Abstract

1412244 Cholesterol dehydrogenase ex Nocar. dia erythropolis BOEHRINGER MANNHEIM GmbH 21 Sept 1972 [22 Sept 1971] 44096/71 Heading C3H [Also in Division G1] A cell free enzyme preparation, free from cholesterol and having cholesterol dehydrogenase activity is obtained by cultivating the micro-organism Nocardia erythropolis in presence of cholesterol as sole carbon source. The cultivation is preferably carried out by inoculating the micro-organism into a salt-cholesterol medium, culturing, harvesting by centrifugation and then suspending the cells as a paste in a phosphate buffer and disrupting them by sonication, deep freezing or expressing them through a small hole. The enzyme is used to assay cholesterol in serum by virtue of its conversion to cholestenone (estimated) quantitatively by U.V. spectroscopy) (see Division G1).
