Abstract

1401238 α-Macroglobulin UNIVERSITY OF STRATHCLYDE 22 Jan 1974 [25 Oct 1972] 49122/72 Heading C3H [Also in Division A5] A pregnancy associated α-macroglobulin of approximate M.W. 506, 500 and the following amino acid composition (in residues of amino acid per 1000 residues) :- and the following carbohydrate composition is per cent by weight:- may be obtained by fractionating human pregnancy blood serum. The fractionation comprises the steps of:- (a) removing #-lipoproteins from the serum, (b) selective precipitation of unwanted compounds, (c) a first dialysis, (d) a first chromatographic separation, (e) a second dialysis, (f) a second chromatographic separation, (g) selective precipitation, (h) gel filtration, (i) selective precipitation, (j) a third dialysis, (k) isoelectric focusing, (1) a fourth dialysis, (m) a third chromatographic separation, a fraction relatively rich in the desired α-macroglobulin being recovered at each of the steps (a)-(1) and passed to the next.
