Abstract

1388719 T-globulin and anti-serum therefor HADASSAN MEDICAL RELIEF ASSOCIATION Inc 1 Sept 1972 [2 Sept 1971] 40595/72 Heading A5B [Also in Division G1] The invention comprises pure T-globulin, i.e. a globulin fraction of M.W. 115,000-170,000 detectable in the blood of pregnant women and cancer sufferers, which when precipitated from serum and subjected to electrophoresis as below migrates towards the positive electrode at 5mm/hr. The electrophoresis is carried out in an agar block measuring 75mm (between the electrodes) x 25mm x 4mm, the current is 10mA, the voltage gradient is 4.5 volts/cm and the agar is buffered at pH 8.6. In preparing T-globulin, blood serum from cancer-sufferers and for pregnant women is treated with saturated ammonium sulphate to precipitate the globulin, from which the appropriate molecular weight fraction is collected as follows: (a) The globulin is dissolved in water (b) a precipitate is formed by addition of perchloric acid, (c) the precipitate is washed with neutral saline, (d) the precipitate is extracted with saline at pH 7.8-8.2 to dissolve out the T- globulin. T-globulin anti serum may be prepared by injecting T-globulin and Freund's complete adjuvant into a mammal. The antiserum may be purified by electrophoresis or by precipitation with perchloric acid followed by re-solution at pH 7.8-8.2. It may further be purified by treating it with normal blood serum to precipitate out all the anti-bodies except anti-T-globulin.
