Abstract

1375836 Indole derivatives JOHN WYETH & BRO Ltd 10 Feb 1972 [18 Feb 1971] 4950/71 Addition to 1218570 Heading C2C Pharmacologically active indole derivatives of the formula wherein R<SP>1</SP> is H, C 1-6 alkyl, C 7-10 aralkyl or aroyl; R<SP>2</SP> is H, C 1-6 alkyl or aryl; R<SP>3</SP> is H, halogen, OH, C 1-6 alkyl or C 1-6 alkoxy; R<SP>4</SP> is H, 'halogen or C 1-6 alkyl; R<SP>5</SP> is aryl (including heteroaryl)-, C 1-6 alkoxy, aryloxy, C 7-10 aralkyl, C 7-10 aralkyloxy, diaryl-C 1-6 alkyl or C 5-7 cycloalkyl; A is C 1-4 alkylene; Z is an oxo (=O) group; and the formula is a 1,2,3,6-tetrahydropyridine or piperidine ring, substituted by NHCZR<SP>5</SP> and R<SP>4</SP> with the proviso that Z may be two hydrogen atoms when R<SP>5</SP> is aryl and the ring is piperidine; are prepared by reacting compounds of the formula with the corresponding 1,2,3,6-tetrahydropyridine or piperidine derivatives in the presence of catalysts. The compounds thus obtained containing a 1,2,3,6-tetrahydropyridine ring may be hydrogenated to the corresponding compounds containing a piperidine ring, or compounds thus obtained in which R<SP>1</SP> is H may be alkylated, aralkylated or aroylated. 4-Cyclohexanecarboxamidopiperidine is obtained by hydrogenating 1-benzyl-4-cyclohexanecarboxamidopiperidine resulting from the reaction between cyclohexane carbonyl chloride and 4-amino-1-benzylpiperidine, obtained by hydrolysing 4 - benzamido - 1 - benzylpiperidine. By similar processes the following intermediates and starting materials are obtained:-1- benzyl - 4 - (3,4 - methylenedioxybenzamido)- piperidine, 4 - (3,4 - methylenediozybenzamido)- piperidine, 1 - benzyl - 4 - (3 - methozybenzamido)piperidine, 4 - (3 - methoxybenzamido)- piperidine, 1 - benzyl - 4 - (4 - methylbenzamido)- piperidine and 4-(4-methylbenzamido)piperidine. Reference has been directed by the Comptroller to Specification 1,273,563.
