Abstract

1364924 Estriol conjugates S J GROSS 4 June 1971 [11 June 1970 16 Nov 1970] 7823/74 Divided out of 1364921 Heading C2U [Also in Divisions C3 C4 A5 and G1] The invention comprises an antigen having a purity of at least 98% and being of formula wherein "carrier" is an immunogenic carrier (e.g. a protein, polysaccharide, polypeptide or glycoprotein); Ar is an optionally substituted arylene or divalent aromatic heterocyclic radical to which the carrier is linked through a connective group (e.g. -NH-CO-); and E is an estriol radical attached to the azo group at position 2 or 4. Specified protein carriers are bovine serum albumin, keyhole limpet hemocyanin and human gamma globulin. The antigen generally comprises a plurality (e.g. 20 or 25) of Ar-N=N-E moieties coupled to a single carrier molecule. The above antigens are prepared from estriol by reaction with diazotized HO 2 C-Ar-NH 2 to give which is chromatographed to separate the 2- and 4-azo isomers from each other and from the 1-azo by-product. The 2- or 4-isomer is then coupled to the carrier in known manner, e.g. by reaction with a carbodiimide or thionyl halide to give or (wherein R 1 and R 2 are each alkyl, cycloalkyl, aryl, alkenyl, cycloalkenyl or heterocyclic) which is then reacted with the carrier to give the desired antigen which is purified, e.g. by dialysis. Reduction of the above HO 2 C-Ar-N = N-E intermediates, e.g. with NaBH 4 , yields a mixture of fluorescent compounds of formula and which may be separated by chromatography and extraction techniques. The above antigens may be made up with diluents into injection solutions.
