Abstract
1359643 Controlled release medicament CONTROLLED MEDICATIONS Ltd 28 Sept 1971 [28 Sept 1970] 46086/70 Heading A5B Shaped, orally administrable medicinal preparations (such as tablets) having extendedrelease characteristics with a 1 to 12 hour release pattern in the gastrointestinal tract comprise a solidly-compacted, void-free body of medicament-solid excipient-binder granules, the binder comprising a pharmaceutically acceptable dicarboxylic acid-monocarboxylic acid ester of cellulose, e.g. cellulose acetate phthalate, the body also including a calcium or magnesium soap in an amount of up to 10% by weight based on the combined weight of the medicament and excipient, may be prepared by forming a mixture of a medicament with a solid excipient, the mixture having a particle size of #100 mesh, granulating the mixture with a binder, and compressing the dry product, a calcium or magnesium soap being added in one or more stages to the mixture and/or granulated mixture prior to the compression step. The calcium or magnesium soap may be calcium or magnesium palmitate, stearate or oleate; the excipient may be a sugar such as lactose, sucrose, dextrose or dextrose monohydrate or may be starch; and the medicament may be a steroid such as ethisterone; a tranquilliser; a diuretic; a sedative; an antibiotic; a vitamin; or an analgesic. The binder may be employed in solution in an organic solvent and the binder may additionally contain up to 10% by weight (based on the total weight of the binder) of a pH-insensitive parmaceutically acceptable binder, e.g. ethyl cellulose.
