1346972 Gatavalin KAYAKU ANTIBIOTIC RESEARCH CO Ltd 10 Oct 1972 46673/72 Heading C2A The new antibiotic gatavalin having a molecular weight of about 2000, a melting point of 245 C.-248 C., an elementary analysis C 51À19%, H 8À19%, N 14À16%, and O 26À46% by difference, a positive reaction to biuret and tert.- butyl hypochlorite, a negative reaction to nin. hydrin, Fehling and Molish; containing 1 mol. glutamic acid, 1 mol. aspartic acid, 2 molalanine, 3 mol. valine and 3 mol. threonine; 23 having a specific rotation [α] +22À4(C:6À5% D in 50% methanol); bioautography on the agar plate seeding St. aureus, n-butanol : acetic acid: water (3 : 1 : 1 by volume) Rf is 0À9 to 0À95; showing end absorption in U.V. and having peaks at 3300, 2900, 1740, 1620, 1520, 1220 and 1150 cm. 1 in IR absorption spectrum (KBr), is prepared by cultivating a gatavalin-producing strain of Bacillus polymyxa var. colistinus KOYAMA, ATTCC No. 21830 in a nutrient medium under aerobic conditions and isolating and recovering the gatavalin from the cultured broth. Gatavalin is isolated and recovered from the cultured broth by acidifying at pH 2 to 5 the culture broth or an aqueous suspension of the cells separated from the culture broth, and thereafter heating, extracting the gatavalin with an organic solvent which is immiscible with water and capable of dissolving the gatavalin, e.g. butyl alcohol, isoamyl alcohol or chloroform, and then precipitating the gatavalin by the addition of a gatavalin-insoluble solvent, e.g. ether, ethyl acetate, butyl acetate or acetone.
