Abstract

1346319 Preserving boar sperm H W SCHAUMANN 30 Nov 1971 [15 July 1971] 55483/71 Heading A2D A method of preserving boar sperm comprises collecting the ejaculate, inactivating said ejeculate, centrifuging and separating the sediment which is then diluted and frozen down to -180 to -200‹C. When required for use the frozen ejaculate is thawed by being placed in a diluent having a temperature of 40 to 50‹C, and then mixed with a solution which mobilizes the spermatozoa. The mobilized spermatozoa demonstrate effective forward motion for at least a four hour period at 39‹C and have a mobility of at least 50% and a keeping quality of at least four hours at 39‹C. The mobilized solution is then applied to the uterus. The inactivating solution may be an aqueous solution of glucose and a buffer system, preferably a solution containing glucose, sodium citrate, sodium bicarbonate, antibiotics, ethylene daimine tertraacetic acid and, potassium chloride. The solution used to dilute the sediment obtained from the centrifuged sperm may be an aqueous solution of fructose, egg-yolk, citric acid, glycerine and a buffer solution. The mobilizing solution preferably comprises sodium salts, a potassium salt, a buffer system and a sulfonamide. Freezing may be carried out by maintaining the ejaculate sediment and diluent at 5‹C for 1“ to 1¢ hours and then causing the sediment and diluent to drip on to dry ice so that pellets are formed and then surrounding the pellets by liquid nitrogen.
