Abstract
1344340 Plasma fractionation STATENS BAKTERIOLOGISKA LABORATORIUM 28 May 1971 [2 June 1970] 17842/71 Heading A5B Blood is fractionated, especially to produce IgG, by the following steps: a) removal of corpuscles and other particles to give plasma, b) precipitating globulins from the plasma by addition of polyethylene glycol (M.W. c. 6000) to a concentration of 10-15% by weight of pH 6.5-8.0, c) removal of the precipitate and, if desired, extraction of albumin from the remaining solution, d) extracting the precipitate with 0.05M sodium acetate/acetic acid buffer at pH 5.8 (IgG dissolved, IgM and fibrinozin remain undissolved and are removed), e) adsorbing the solution from d) on a cation exchanger (e.g. carboxy methyl dextran) at pH 5.8 and eluting with 0.2M NaCl in 0.05 phosphate buffer at pH 7.5-8.0, f) reprecipitating the eluate with polyethylene glycol at pH 6.5-8.0 and dissolving the precipitate in 0.2M phosphate buffer at pH 6.6, g) adsorbing all the globulins other than IgG from the solution from f) on an anion exchanger, e.g. DEAE- dextran at pH 6.6, h) cooling the resulting IgG solution to -0.3‹C and precipitating with ethanol (25% concentration) at -5 to -10‹C, i) separating and suspending the precipitate and washing it with 25% ethanol at pH 7.0 at -5 to -10‹C, j) removing solids by centrifugation and dissolving in glycine, k) sterilefiltering the solution and freeze-drying.
