Abstract
1331435 Pyridylmethylcarbonyloxy-blocked amine derivatives RHONE-POULENC SA 4 Aug 1970 [5 Aug 1969 9 June 1970] 37633/70 Headings C2A and C2C [Also in Division C3] Polyfunctional compounds containing one or more amino radicals with at least one of said radicals protected by a group: (where Py is pyridyl or pyridyl N-oxide, optionally substituted by a methyl radical) are prepared by reacting the said polyfunctional compound with Py-CH 2 -OCO-X (where X is a radical which activates the carbonyl group, e.g. halogen, azido or the radical OT and T is phenyl optionally substituted or a radical derived from a heterocyclic compound). Polyfunctional compounds may be amino acids, peptides, cyclopeptides, polymyxin or colistin or their derivatives. The free peptides are prepared by removing the blocking group by hydrogenolysis, Na and ammonia or by selective enzyme hydrolysis, e.g. of a penta-protected colistin to a tri-protected colistamine. The following starting materials are also prepared: 3- and 4-hydroxymethylpyridine-N-oxides 2 - hydroxy - 6 - methyl - pyridine, 3 - and 4 - pyridylmethyl p - nitrophenyl carbonate, (6 - methyl - 2 - pyridyl) methyl p - nitrophenyl carbonate, 3 - pyridylmethyl 2,4,5 - trichlorophenyl carbonate, (3 - pyridyl - N - oxide)- methyl p - nitrophenylcarbonate, (6 - methyl- 2 - pyridyl - N - oxide)methyl p - nitrophenylcarbonate, (4 - pyridyl - N - oxide) methyl - pnitrophenyl carbonate, 3-pyridyl-N-oxide) methyl 2,4,5-trichlorophenyl carbonate or p-nitrophenyl carbonate.
