1321991 Guanidine derivatives WELLCOME FOUNDATION Ltd (Burroughs Wellcome & Co (USA) Inc) 22 June 1970 [23 June 1969 4 Dec 1969] 31684/69 and 59232/69 Heading C2C Novel acid addition salts of N-p-methoxybenzyl - N<SP>1</SP>,N<SP>11</SP> - dimethylguanidine are prepared by (a) reacting a guanidine with a compound BX where one of A and B is the p-methoxybenzyl group and the other is the methyl group and X is a sulphonate group or halogen atom, or (b) reacting an appropriate amine or salt thereof with an S-substituted isothiourea or salt thereof, an O-substituted isourea or salt thereof, an isocyandihalide, an imidocarbonate or imidothiocarbonate, a diimide or a formamidine substituted by an unsaturated heterocyclic group containing at least two nitrogen atoms in the ring, one of which is attached to formamidine structure, or (c) salifying the free base or converting one acid addition salt into another. Intermediates.-The following compounds are prepared in the examples: N-p-methoxybenzyl- N<SP>1</SP>,S - dimethylisothiourea hydriodide, N - pmethoxybenzyl - N<SP>1</SP> - methylthiourea, p - methoxybenzonitrile, p - methoxybenzylamine hydrochloride, N - p - methoxybenzyl diethyl iminocarbonate, diethyl chloroiminocarbonate, diethyl bromoiminocarbonate, and diethyl iminocarbonate. Pharmaceutical compositions for oral or parenteral administration comprise a non-toxic acid addition salt of the invention together with a pharmaceutical carrier. The compositions have anti-arrhythmic activity.
