Abstract
1314263 Determination of blood components MALLINCKRODT CHEMICAL WORKS 22 April 1970 [1 Ma,y 1969 13 Oct 1969] 19391/70 Heading BIX Blood components which can be bound by serum protein, for example thyroxine or triiodothyronine, or iron are determined by a method comprising preparing a liquid sample containing blood serum protein and a predetermined amount of a radioactive tracer compound capable of being bound by the serum protein and by an ion exchange resin, adding to the sample a thin strip of an ion exchange resin membrane and maintaining sample and strip in contact for a predetermined time, separating the membrane from the sample and determining the relative amounts of the labelled compound bound to the serum protein in the sample and to the membrane. The blood serum protein may be globulin, albumin or prealbumin. The radioactively labelled compound may be I<SP>125</SP>-thyroxine or I<SP>131</SP>-L-triiodothyronine for determination of the total thyroxine content of serum or the throxine binding capacity of serum protein, or ferric (Fe<SP>59</SP>) ammonium citrate for determination of the iron-binding capacity of serum proteins. The membrane may be a strip, sheet or film of solid hydrous gel of thickness 5 mils to 50 mls and may be comprise anionic or cationic exchange resin. Exemplified is a membrane comprising a vinyl copolymer carrying quaternary ammonium groups, the membrane being reinforced with loosely woven fibres of a co-polymer of vinyl chloride and acrylonitrile. A reagent for use in the determination of thyroxine content is prepared by diluting blood serum with barbital buffer pH 8.6, passing the diluted serum through an anion exchange column, which adsorbs about 80% of the thyroxine, and adding radioactive thyroxine to the filtered eluate.
