Abstract

1300291 Haematology control fluid; stabilising fowl red cells PFIZER Inc 17 July 1970 34665/70 Heading B1X Fowl red blood cells are stabilized by a process comprising mixing an 8% suspension of the cells in 0.45% saline with an equal volume of 1% mercuric chloride in 0.45% saline, heating the resulting mixture at 37‹C for about 19 hours, centrifuging and resuspending the cells as a 4% suspension in saline, combining with the cell suspension an equal volume of a 0.0025% solution of tannic acid in 0.45% saline buffered at pH 7.2, incubating for 30 to 60 minutes at about 50‹C, centrifuging, incubating at 0.01 to 20% suspension of the cells in 5.0% glutaraldehyde in an isotonic solution buffered at pH 7.2 at 37‹C for 16-20 hours and washing the cells 3 or 4 times with the buffered isotonic solution, separating the cells by centrifugation after each washing. A hematology control fluid comprises a suspension of fresh human red blood cells and the tanned fowl red blood cells prepared as above in a volume of anticoagulant-containing serologically compatible human plasma medium which is approximately equal to the packed volume of the human cells, the concentration of the fowl cells being 5,000-10,000 per cubic millimeter. The fowl cells may be goose, chicken, duck or (preferably) turkey cells. Reference has been directed by the Comptroller to Specifications 1,131,690 and 1,215,662.
