Abstract

1300128 Cephalosporin-γ-lactams ROUSSELUCLAF 19 Oct 1970 [17 Oct 1969] 49516/70 Addition to 1155493 Heading C2A Novel 7 - acylamino - 3 - aminomethyl - cephalosporanic acid-γ-lactams having the Formula I<SP>1</SP> and salts thereof, wherein X is -NO 2 or -NH 2 and may be at position 4, 5, 6 or 7 of the indole nucleus, are prepared by acylating the 7-amino group of 7 - amino - 3 - aminomethyl - cephalosporanic acid-γ-lactam with a (nitro-indol-3<SP>1</SP>-yl)- acetic acid or an N-acylating derivative thereof and optionally subjecting the product to catalytic hydrogenation to convert the -NO 2 group to -NH 2 . The preferred acylating agent is the acid anhydride formed in situ from the acid and a condensing agent (e.g. dicyclohexyl carbodiimide). The hydrogenation is performed in the presence of palladinized charcoal. In an alternative process, the compounds (I<SP>1</SP>) may be prepared by lactamization of an appropriate 7 - acylamino - 3 - aminomethyl cephalosporanic acid or its ester. Pharmaceutical compositions having antibiotic activity against gram-positive and -negative bacteria, comprise a cephalosporin-γ- lactam of Formula I<SP>1</SP> above, or a therapeutically acceptable salt thereof together with a pharmaceutical carrier. The compositions may be in the form of tablets, capsules, pills, powders, oral or injectable liquid forms, topically applicable forms or suppositories.
