1299580 Hetrocyclic phenylalkylamines LILLY INDUSTRIES Ltd 15 Jan 1970 33625/72 Divided out of 1299172 Heading C2C Novel compounds of Formula L wherein n is 1 or 2; R<SP>2</SP> is an -NH 2 or -NR 2 group where R is a methyl, ethyl or n-propyl group or the two groups R taken together with the nitrogen atom to which they are attached represent a morpholino radical, and R<SP>1</SP> is H<SP>2</SP> or HR where R is as defined above; R<SP>3</SP> is a hydrogen, chlorine, bromine, fluorine or iodine atom or a methyl, ethyl, methoxy, hydroxy, acetyl, trifluoromethyl, -SO 2 CH 3 , -SOCH 3 , nitro, amino, dimethylamino, benzylmercapto or methylthio group; R<SP>4</SP> is hydrogen, and in no more than one instance where it occurs on the alkylene chain it may also be a methyl, ethyl, n-propyl or -CH(OH)CCl 3 group; and A is a 1 -adamantylamino radical or a heteryclic group of the formula wherein Y is #CH- and X is #N.CH 3 , or Y is a nitrogen atom and X is #N.CH 3 , a methvlene group, an oxygen atom or a sulphur atom; and x and y are selected from zero or integers from 1 to 6 such that the sum of x and y is 3, 4, 5 or 6; and the pharmaceutically acceptable acid addition salts thereof are prepared either by reduction of the nitrile of formal L above wherein -CR 1 R 2 is -C#N or by one of the following reaction schemes wherein the symbols have the above meanings and B is a nitrile, ester or carboxylate group. Starting material wherein A is piperidino are prepared by reaction of the compound where A is amino with 1,5-dibromopentane. 2-Piperidimobenzaldehye is prepared by reaction of piperdine with 2-flaorobenzaldihyde. 3 - Chloro - 4 - piperidinobenzaldehyde is prepared by action of sodium nitrite and cuprous chloride on the acidified 3-amino compound prepared by reduction of 3-nitro-4- piperidino benzaldehyde obtained by condensation of 4-chloro-3-nitrobenzaldehyde and piperidine. Pharmaceutical compositions in conventional forms for oral, rectal, parenteral and topical administration having anti-inflammatory activity comprise an above novel compound and a carrier or diluent.
