Abstract

1,273,618. Estradiol derivatives. WARNERLAMBERT CO. 9 Dec., 1969 [11 Dec., 1968 (3); 25 July, 1969 (2)], No. 60088/69. Heading C2U. The invention comprises compounds of formula (wherein R is alkanoyl, phenylalkanoyl, cycloalkylalkanoyl of up to 10 carbon atoms or is a benzoyl radical optionally substituted by C 1-5 alkyl, C 1-5 alkoxy, cycloalkyloxy, nitro or halo; and n is an integer from 1 to 23) and their preparation from the corresponding 17#-ols by etherification with a dialkyl acetal or alkyl enol ether of a C 7-29 cycloalkanone. The o-, m- and p-chlorobenzoates, -fluorobenzoates, and -methoxybenzoates and the p-cyclopentyloxybenzoate, p-toluate and p-nitrobenzoate of estrone and estradiol are prepared by esterification of estrone followed, when required, by reduction of the 17-oxo group. The inventive compounds are said to possess uterotrophic, estrogenic and contraceptive activities, and may be made up with carriers into pharmaceutical compositions for oral and parenteral administration.
