Abstract

1,214,891. Thyrocalcitonin. ELI LILLY & CO. 1 March, 1968 [2 March, 1967], No. 10112/68. Heading A5B. Thyrocalcitonin having a significant serumcalcium-lowering activity in rats at a dose level of 5γ per kilo of rat body weight is prepared by extracting mammalian thyroid tissue with an acidic mixture of from 25 to 85 percent by volume of a water miscible organic solvent and water, e.g. acidic aqueousethanol, at a pH below 4, preferentially precipitating inactive proteinaceous materials from the extract by the addition of 1 to 3 volumes of watermiscible organic solvent per volume of extract or by adjusting the pH of the extract to about 6.5, separating the precipitated inactive proteinaceous material from the extract, precipitating thyroalcitonin from the extract by the addition of 5 to 10 volumes of a watermiscible organic solvent per volume of extract, and finally separating and purifying the thyrocalcitonin thus precipitated. The water-miscible solvent may be dioxane, dimethylformamide, acetone, methanol, isopropanol or ethanol and the acid may comprise hydrochloric, phosphoric, formic, acetic or sulphuric acid. The thyrocalcitonin may be purified by gel filtration or chromatography over a polydextran, optionally followed by lyophilization.
