The concentration of a substance in solution is measured by treating the solution with a reagent that reacts with the substance to vary the concentration of oxygen in the solution in proportion to the initial concentration of the substance and measuring the change or rate of change in the oxygen concentration using an oxygen electrode. As described with reference to Fig. 1, the reagent is introduced into chamber 1 through inlet 3, and may be pumped continuously through the cell for initially washing the chamber. The solution under test is added through an injecting lance 13. An electrolytic cell for measuring oxygen concentration comprising a platinum electrode 11, glass coated at 10, and silver/silver chloride reference anode 12 in a buffer solution containing phosphate and chloride ions is separated from the sample by an oxygen-permeable membrane 7. Stirring is by a magnet 15 on a plastic covered magnetizable slug 17. In an alternative arrangement, the membrane 7 is omitted so that the oxygen electrode is in direct contact with the solution under test. In this case the electrode is vibrated or rotated to maintain a continuously renewed contact between it and the surrounding liquid. Many examples of substances that can be measured and appropriate reagents are given. Glucose can be measured, using a 1% glucose oxidase solution saturated with air as the reagent, the slope of the recorded oxygen concentration being a measure of the concentration of glucose in the sample. If desired, a 0À1% alcohol or peroxidase and an aromatic amine or phenol may be added to the reagent to preserve stoichiometry in the presence of catalase. Galactoze, uric acid, ascorbic acid (vitamin C), amino acid, amine, diamine, xanthine, aldehyde, lipid and phenolic oxidases can be used as reagents or may be measured themselves. In general, any reaction which produces a molecule for which an oxidase or dehydrogenase exists can be measured by measuring oxygen consumption. Hydrogen peroxide can be measured, by catalytic conversion to water and oxygen. Catalase can be measured by measuring the rate of oxygen liberation from hydrogen peroxide or an alkyl hydroperoxide.
