Abstract
1,195,932. Quinazolinylureas. CHAS. PFIZER & CO. Inc. 9 May, 1968 [2 Feb., 1968], No. 22133/68. Heading C2C. Novel quinazolinylureas of the formula wherein R 3 is H or C 1-6 alkyl; R 4 and R 5 are H or C 1-3 alkoxy, at least one of R 4 and R 5 being alkoxy; and R 1 and R 2 are the same or different and are H, C 1-5 alkyl, C 3-5 alkenyl, C 2-5 hydroxyalkyl, phenyl, benzyl, phenylethyl, 2-furfuryl, 2,2,2-trifluoroethyl, or C 3-8 cycloalkyl, or R 1 and R 2 and the N atom to which they are attached form a morpholino, 1-azacycloheptyl, 1-azacyclooctyl, or a 4-Y-piperazino, wherein Y is H, CF 3 , C 1-5 alkyl, C 2-5 hydroxyalkyl, C 2-7 - alkanoyl, allyl, propargyl, 2-methyallyl, phenyl, benzyl, benzoyl, halobenzoyl or halophenyl, where halo is chloro or bromo, trifluoromethyl. phenyl, methoxyphenyl, methylphenyl, methylbenzoyl, methoxybenzoyl, trifluoromethylbenzoyl, furoyl, benzofuroyl, thenoyl, pyridinecarbonyl, 3,4,5-trimethoxybenzoyl, carboxylic acid C 1-6 alkyl or C 3-6 alkenyl ester or 4-X-4-X<SP>1</SP>- piperidino where X<SP>1</SP> is H or phenyl, and X is H, C 1-5 alkyl, C 1-4 alkoxy, OH, C 2-5 hydroxyalkyl, phenyl or benzyl when X<SP>1</SP> is H, or X is carboxylic acid C 1-5 alkyl ester when X<SP>1</SP> is phenyl; and acid addition salts thereof are prepared either by reacting the corresponding 4-aminoquinazolines, provided R 1 and R 2 are not hydrogen and do not contain hydroxyl groups, with R 3 NCO or, for compounds where R 3 is hydrogen, with NH 4 NCO, KNCO and NaNCO in the presence of a mineral acid, or by reacting amines of the formula HN(R 1 )R 2 with the corresponding 1-(2- chloro - 4 - quinazolinyl) - 3 - alkylurea or (2- chloro-4-quinazolinyl)-urea, which are obtained by treating the corresponding 2-chloro-4-aminoquinazolines with R 3 NCO and NH 4 NCO, KNCO and NaNCO in the presence of a mineral acid respectively. Pharmaceutical compositions, having hypotensive activity, and suitable for oral or parenteral administration, contain the above novel quinazolinylureas or pharmaceutically acceptable acid addition salts thereof as the active ingredient.
