Abstract

1,194,256. Immunological indicator comprising microbial cells. MILES LABORATORIES Inc. 19 June, No. 28181/67. Heading B1X. An immunological indicator comprises microbial cells to which are attached coupling agent molecules by covalent bonds. The attached coupling agent molecules each have at least one non-bonded group which can form a covalent bond with antigens, giving an "indicator system". The microbial cells may be bacterial, fungal or protozoal cells with a maximum dimension of 0.2 to 10 microns, and may 'be treated with formaldehyde or phenol and optionally stained before use. The coupling agent may be bis diazobenzidine (BDB), bis diazobenzidine disulphonic acid, tetra-azo-pphenylene-diamine, difluorodinitrobenzene, a carbodiimide, toluene diisocyaniate, cyanuric chloride, dichloro-s-triazine, N-t-butyl-5-methyl -isoxazolium perchlorate. Indicator systems exemlified are Brucella abortus-BDB-HCG; E. coli-BDB-HCG Saccharomyces cerevisiae- BDB-HCG; E.Coli-N,N'-dicyclohexyl carbodiimide-HCG; E. coli-BDB-human serum albumin (HSA); E. coli-BDB-horse gamma globulin; E.coli-N-t-butyl-5-methylisoxazoliuma perchlorate-HSA; the same indicator coupled to horse gamma globulin; each of Bacillus subtilis, Bacillus pumilis, Lactobacillus leichmannii, Pseudomonas fragi coupled to BDB and to each of HSA, horse gamma globulin and human insulin; other components are mentioned. The indicator systems may be used in chromatography type tests, tube tests, slide or plate-tests. Test devices for chromatography type tests for antigens comprise bibulous materials carrying the residue resulting from either (a) the deposition of an indicator system as above, made from the antigen to be detected, and a superimposed deposition of the homologous antibody, or (b) the deposition of the immune aggregate formed from the indicator system and the homologous antibody. The device may be developed with a urine sample under test or a spot of the urine sample may be placed on the spot of immune aggregate and the device developed with saline. If the immune aggregate dissociates in the presence of the sample, the indicator system migrates, and if the cells were stained the migrating material forms coloured streaks.
