Abstract

1,169,183. Density gradient electro-phoresis apparatus. INSTRUMENTATION SPECIALTIES CO. Oct.3, 1966 [May 25, 1966], No.44084/66. Heading C7B. Apparatus for density gradient electro-phoresis comprises a vertically positioned tube of electrical insulating material for receiving density gradient liquid, means including a reversible pump for pumping a displacing liquid into and out of the tube through the lower end thereof to vary the position of the density gradient liquid within the tube, and means establishing an electric field across axially-spaced points within the tube comprising electrodes connected to a source of D.C. voltage and immersed in buffer solutions contained in chambers having communication with the tube at the axially spaced points through openings covered by semi-permeable membranes. In the embodiment shown the tube for containing the density gradient liquid comprises tubes 11, 11' positioned end to end and separated by a quartz ring 21 at opposite sides of which are positioned a lamp 16 and photo-cell 24, respectively, forming a photo-metric scanning system. By pumping displacing liquid into and out from the tube via fitting 67 the fractionated zones of the sample may be passed by said scanning system and the light absorption, and hence the concentration of the fractionated zones, may be measured, which scanning process may be repeated as often as desired. The upper end of the tube 11 abuts a tube 27 having appertures 38 covered by semipermeable membranes, which appertures connect with chambers 31 and hence with trough 34, the chambers 31 and trough 34 containing a buffer salt solution wherein is positioned an electrode 36. Similarly the lower end of the tube 11' abuts tube 47 having apertures 51, covered by semipermeable membranes, contacting openings 52 and 53, an inner trough 56 and an outer trough 60, the openings 52, 53 and the troughs 56, 60 containing a buffer salt solution, the troughs being inter-connected via openings 57 covered by semi-permeable membranes 58, and the outer trough containing a counter-electrode 46. The tubes 27, 11, 11' and 47 are contained within the concentric tubes 40, 44, 64 which provide a cooling jacket, cooling fluid being passed therethrough between hose nipples 42 and 65, the cooling jacket spaces being inter-connected by passages 45 and 66. The density gradient fluid may be passed into the tube via the lower opening 67 or may be pipetted therein via the upper opening 43.
