Abstract

1,141,735. Naphthoquinone derivative. CHAS. PFIZER & CO. Inc. 15 March, 1967 [23 Nov., 1966], No. 12259/67. Heading C2C. [Also in Division A5] The compound lapachol (2-hydroxy-3-(3- methyl-2-butenyl)-1,4-naphthaquinone is obtained by extracting the roots or heartwood of Stereospermum suaveoleus three times with cold ligroin and concentrating the combined extracts. Other solvents, e.g. benzene, CHCl 3 , ethyl acetate, MeOH or aqueous Na 2 C0 3 may be used. If MeOH is employed, the extract is concentrated and shaken with benzene and water. The benzene layer is concentrated and chromatographed on silicic acid/cellulose or magnesium silicate by elution with benzene, the yellow band of lapachol being recovered and recrystallized.
