Abstract
<PICT:1119013/C2/1> A new antibiotic, dermadin, is produced by culturing a dermadin-producing strain of Trichoderma viride in aqueous nutrient medium under aerobic conditions. It is an acidic substance containing 60.18% carbon, 4.09% hydrogen, 26.78% oxygen, and 7.56% nitrogen, having a molecular weight of 177 and empirical formula C9H7NO3, and being soluble in chloroform, methylene chloride, methanol, ethanol, ethyl acetate, and water. It has an ultra-violet absorption peak at 223 mm and the infrared spectrum shown above. It has an optical rotation of [a ]25D = + 138 in 0.88% solution in 50% aqueous ethanol. It is bacteriostatic to gram positive and gram negative bacteria, and inhibits the growth of fungi, algae and human epidermoid carcinoma cells in tissue culture. The strain of micro-organism used is Trichoderma viride NRRL 3153. Culture conditions are preferably 25 DEG to 30 DEG C., at pH 6 to 8, the vegetative form of the organism being inoculated into a nutrient medium containing assimilable nitrogen and carbon sources, such as casein digest, meat and bone scraps, milk solids, peptone liquors or distiller's solubles for a nitrogen source, and sugar, starch, dextrin or glycerol to supply carbon. After culture, preferably submerged culture, the antibiotic may be obtained by solvent extraction, with, e.g. ethyl acetate, followed by water extraction of the solvent solution at neutral pH. The aqueous extract may be freeze dried, or further purified by use of ion exchange procedures, or by adsorbing on such agents as silicic acid, decolorizing charcoal or decolorizing resins, followed by elution with solvent. It may be crystallized from methylene chloride by addition of hexane. Dermadin forms salts with metals, such as alkali metals, and organic bases including physiologically active bases (see Division A5).
