Abstract

Anti-coagulant material is prepared from the blood-sucking insect Rhodnius prolixus stal. by centrifuging a suspension in a saline medium of the homogenised insect, to remove tissue debris, treating the supernatant with a protein precipitant, extracting the resulting precipitate with a saline medium, and recovering the saline extract containing the required material. The preferred starting material is the excised gut of the insect; the protein precipitant may be acetone, methyl ethyl ketone, or other organic liquids, or inorganic salts may be used. The saline extract may be concentrated, if required to dryness, in a vacuum dessicator at room temperature. A further purification by heating the extract to 80 DEG C. for ten minutes, to precipitate unwanted protein, may be performed before drying. The product has anti-thrombin activity similar to heparin, and a molecular weight of 100,000 to 200,000.
