Abstract
In the propagation of animal cells, in which the cells are suspended in an aqueous culture medium containing or being supplied with nutrients or additives known to be necessary or useful for the sustenance or multiplication of cells, the pH of the medium is kept between 7.3 and 7.8. For the control of the pH, the medium is contacted with a non-poisonous gas (e.g. by flowing the gas across the top of the medium) which contains carbon dioxide at a partial pressure higher than the equilibrium pressure corresponding to the carbon dioxide content of the medium should the pH of the medium rise above a desired or defined maximum pH, and a non-poisonous gas which contains carbon dioxide at a partial pressure below the above-mentioned equilibrium pressure is sparged through the medium should the pH of the medium fall below a desired or defined minimum pH. Alternatively, there is sparged through the medium a non-poisonous gas containing an adjusted amount of carbon dioxide for the regulation of the pH. The gas may consist of air, air diluted with an inert gas, or a mixture of non-poisonous gases preferably containing some oxygen. Non-poisonous anti-foam agents may be added to the medium to avoid frothing. Examples are directed to the propagation of cloned baby hamster kidney cells and to growing strains of foot-and-mouth disease viruses in the cells for the production of vaccine.ALSO:Strains of foot-and-mouth disease viruses are grown in cloned baby hamster kidney cells propagated in suspended culture in an aqueous culture medium regulated to a pH within the range 7.3 and 7.8.
