Abstract

Analysis of an object using a photo-electric microscope involves taking a series of measurements which are stored and then compared with a second series of measurements. In the embodiment described, light from a source (12) passes through a variable monochromator, through the object plane to a photo-cell (3) which is mounted on the microscope tube. Signals corresponding to the intensity incident on the cell pass to an amplifier (5). A beam splitter (11) in the light path and further photo-cell (10) may be provided to compensate for variations in source intensity, by comparing the outputs of the photo-cells in the amplifier (5). In the analysis of the object, a first series of reference values is obtained by varying the wavelength of light incident on a part of the microscope slide not containing the sample, by adjustment of the monochromator. The output of the amplifier (5) is recorded on a tape (7) running in synchronism with the monochromator adjustment. The amplifier output is passed through a converter (6) which may, for example, generate an A. C. signal the frequency of which corresponds to the amplifier output value. The monochromator adjustment is then repeated with part of the object in the light path, the tape now being read out in synchronism so that the reference values previously recorded may be compared, after passing through the converter, with the amplifier output in a comparison device (8), the output of which is indicated or recorded. Instead of a wavelength variation, an extended area of the object may be examined at a constant wavelength. If a plurality of tape tracks are provided, a recording of the measurements on the object may be made on the tape, alongside the reference values. An information track for speech &c. may also be provided. The signals may be converted to digital form for storage.
