Abstract

31 - Phosphoadenosine - 51 - phosphosulphate and/or adenosine-51-phosphosulphate, or salts thereof, are prepared by inoculating micro-organisms belonging to the genus Streptomyces in an aqueous nutrient medium containing sources of assimilable carbon, nitrogen, phosphate and sulphate, incubating the inoculated medium and recovering the desired substances from the culture broth. Examples of carbon sources are starch, dextrin, glucose, sucrose, lactose, maltose and glycerol, examples of nitrogen sources are peptone, yeast extract, yeast, soya bean meal, cornsteep liquor, amino acids, ammonium salts and nitrates, examples of phosphate sources are dipotassium hydrogen phosphate and potassium dihydrogen phosphate and magnesium sulphate is specified as the sulphate source. The pH of the medium is preferably 7 and the incubation is suitably carried out at 20 DEG C. to 37 DEG C. After incubation the desired substances may be separated from the culture broth by acidifying and filtering the broth, treating the filtrate with activated charcoal, and eluting from the charcoal with ammoniacal aqueous ethanol. The eluate obtained may then be treated with a basic anion-exchange resin and adenosine-51-phosphosulphate and 31-phospho-adenosine-51-phosphosulphate are successively eluted with an aqueous solution of formic acid and ammonium formate. The eluates obtained are then freeze-dried. Numerous examples are given of suitable Streptomyces micro-organisms. 31 - phosphoadenosine - 51 -phosphate, adenosine - 51 - monophosphate, adenosine diphosphate, and adenosine triphosphate may also be isolated in minor amounts from the culture broth.
