L’endocan murin a ensuite été concentré par centrifugation en utilisant une membrane semi-perméable (Amicon® Ultra Centrifugal Filters). Une détermination de la concentration finale en endocan a été réalisée avec un kit ELISA spécifique de dosage d’endocan (réf. LIK-1101, DIYEK EndoMark M1, Biothelis, France). Chaque lot d’endocan produit a finalement été contrôlé par la technique de Western Blot pour s’assurer de la présence exclusive de la forme glycosylée, et un dosage de LPS par un test spécifique (LAL test (réf. 50-650U, Lonza, Switzerland)) a été réalisé sur chaque lot pour garantir de l’absence de LPS.The glycosylated murine recombinant endocan was produced, then purified, in transfected CHO cells as previously described [9]. CHO cells were cultured in suspension in serum-free medium (serum-free CHO media (Gibco, Waltham, Massachusetts, USA)) and placed in bioreactors (ref. CL1000 bioreactors (ref. Z688029, Integra Biosciences, Cergy , France)) in order to obtain a cellular supernatant rich in glycosylated murine endocan. After collecting the supernatant, the recombinant endocan was purified on an ion exchange column (HiTrap Q FF (ref. 17-5156-01, GE healthcare, USA)), then on an affinity column coated with anti-murine endocan MEP14 antibody, as previously described [9]. The murine endocan was then concentrated by centrifugation using a semi-permeable membrane (Amicon® Ultra Centrifugal Filters). A determination of the final endocan concentration was carried out with a specific endocan assay ELISA kit (ref. LIK-1101, DIYEK EndoMark M1, Biothelis, France). Each batch of endocan produced was finally controlled by the Western Blot technique to ensure the exclusive presence of the glycosylated form, and a dosage of LPS by a specific test (LAL test (ref. 50-650U, Lonza, Switzerland)) was carried out on each batch to guarantee the absence of LPS.
1.21.2 Animaux et aspect éthiquesAnimals and ethical aspects
Nous avons eu recours à des souris mâles C57Bl/6 Esm1-/- et WT littermate, âgées de 7 – 11 semaines et en reproductions dans notre animalerie. Le protocole de l’étude a été approuvé par la commission d’éthique locale et toutes les expérimentations animales ont suivi les règles de bonnes pratiques éthiques (n° APAFIS#6059).We used male C57Bl/6 Esm1-/- and WT littermate mice, aged 7 – 11 weeks and breeding in our animal facility. The study protocol was approved by the local ethics committee and all animal experiments followed the rules of good ethical practice (no. APAFIS#6059).
1.31.3 Obtention des souris Esm1-/-Obtaining Esm1-/- mice
Pour obtenir les lignées C57Bl/6 Esm1-/-, nous avons inséré des loci de reconnaissance loxP autours des exons 1 et 2 du gène murin Esm1 (Ensembl ID ENSMUSG00000042379), puis, en collaboration avec l’Institut Clinique de la Souris (Strasbourg, France), nous avons transfecté par électroporation un fragment d’ADNc porteur d’une cassette de résistance à la néomycine encadrée par des loci loxP dans des cellules souches embryoniques murines. Nous avons alors obtenu par recombinaison homologue un remplacement des exons 1 et 2 du gène Esm1 par le fragment porteur du gène de résistance à la néomycine. Nous avons ensuite sélectionné les cellules souches embryonnaires transfectée par culture dans du milieu contenant du G418, puis nous avons vérifié la délétion par technique Southern blot. Pour finir, nous avons injecté les cellules embryonnaires déficientes Esm1-/- dans des blastocystes secondairement placés dans l’utérus de souris femelles pseudo gestantes. Nous avons ensuite croisé la progéniture obtenue pour obtenir des lignées de souris déficientes homozygote en exon 1 et 2 du gène Esm1. Enfin, nous avons croisé ces souris avec d’autres souris porteuses d’une délétion Cre pour éliminer le gène de résistance à la néomycine. Pour vérifier la délétion Esm1-/-, un génotypage a été réalisé après extraction d’ADN à partir d’un prélèvement biologique, et une PCR a été réalisée avec le kit REDExtract-N-Amp™ Tissue PCR kit (réf. 254-457-8, Sigma-Aldrich, USA avec les amorces suivantes : 1f (SEQ ID NO : 2 : ATGTCTATGTCAGTCTCCTC) et 1r (SEQ ID NO : 3 : CTCTTGCCAGCCTCTCTTGT) pour générer un fragment de 310 paires de bases pour l’allèle sauvage et 1f et 2r (SEQ ID NO : 4 : CTCCAAAATCCAGAAACCC) pour générer un fragment de 442 paires de bases pour l’allèles KO. We then crossed the resulting offspring to obtain lines of mice homozygously deficient in exon 1 and 2 of the Esm1 gene. Finally, we crossed these mice with other mice carrying a Cre deletion to eliminate the neomycin resistance gene. To verify the Esm1-/- deletion, genotyping was carried out after extraction of DNA from a biological sample, and a PCR was carried out with the REDExtract-N-Amp™ Tissue PCR kit (ref. 254- 457-8, Sigma-Aldrich, USA with the following primers: 1f (SEQ ID NO: 2: ATGTCTATGTCAGTCTCCTC) and 1r (SEQ ID NO: 3: CTCTTGCCAGCCTCTCTTGT) to generate a 310 base pair fragment for the wild allele and 1f and 2r (SEQ ID NO: 4: CTCCAAAATCCAGAAAACCC) to generate a 442 base pair fragment for the KO allele. The samples were then analyzed on agarose gel.

1.51.5 Préparation des osmopompes et implantation sous cutanéePreparation of osmopumps and subcutaneous implantation
Nous avons eu recours à des osmopompes permettant une administration continue de la solution chargée durant 3 jours, à un débit fixe de 1 µL.h-1. Chaque osmopompe de 100 µL a été remplie soit avec 200 µg d'endocan recombinant murin glycosylé (100 µl de solution d'endocan à une concentration finale de 2 mg.mL-1), soit par 100 µL de PBS. Les osmopompes ont ensuite été immergées dans du PBS pour une durée maximale de 4 heures avant leur implantation en sous cutanée. Après avoir anesthésié les souris conformément au protocole défini plus loin, nous avons réalisé une désinfection cutanée puis nous avons implanté les osmopompes en sous cutané. L'anesthésie générale a alors été antagonisée par injection intrapéritonéale de 5 mg.kg-1d'atipamezole.We used osmopumps allowing continuous administration of the loaded solution for 3 days, at a fixed flow rate of 1 µL.h -1 . Each 100 µL osmopump was filled either with 200 µg of glycosylated murine recombinant endocan (100 µl of endocan solution at a final concentration of 2 mg.mL -1 ), or with 100 µL of PBS. The osmopumps were then immersed in PBS for a maximum period of 4 hours before their subcutaneous implantation. After anesthetizing the mice in accordance with the protocol defined below, we carried out skin disinfection and then implanted the osmopumps subcutaneously. General anesthesia was then antagonized by intraperitoneal injection of 5 mg.kg -1 of atipamezole.
1.61.6 Préparation et administration du LPS pour le modèle d'agression pulmonairePreparation and administration of LPS for lung insult model
Le LPS utilisé dans notre protocole (E. Coli O111 : B4, Sigma-Aldrich, USA) a été préparé en conditions stériles. Pour chaque souris, 10 mg.kg-1de LPS ont été dilués dans du PBS pour obtenir un volume final de 50 µL. Pour le protocole d'agression aigue pulmonaire induite par le LPS, les souris ont été placées sous anesthésie générale par injection intrapéritonéale de 75 mg.kg-1de kétamine et de 0,5 mg.kg-1demedetomidine, puis, la solution contenant le LPS a été administrée par instillation intratrachéale. L'anesthésie générale a ensuite été antagonisée comme précédemment décrit.The LPS used in our protocol (E. Coli O111: B4, Sigma-Aldrich, USA) was prepared under sterile conditions. For each mouse, 10 mg.kg -1 of LPS was diluted in PBS to obtain a final volume of 50 µL. For the LPS-induced acute pulmonary insult protocol, the mice were placed under general anesthesia by intraperitoneal injection of 75 mg.kg -1 of ketamine and 0.5 mg.kg -1 of demedetomidine, then, the solution containing LPS was administered by intratracheal instillation. General anesthesia was then antagonized as previously described.
1.71.7 Évaluation clinique quotidienne et mesure non invasive de l'oxymétrieDaily clinical assessment and non-invasive measurement of oximetry
Dans chaque protocole, un examen clinique quotidien des souris a été réalisé afin d’évaluer la viabilité de la souris et de mesurer son poids. Un enregistrement continu non invasif de l'oxymétrie transcutanée, du rythme cardiaque ainsi que de la fréquence respiratoire a été réalisé sur souris vigile quotidiennement par le dispositif MouseOx® Plus device (Starr Life Sciences Corp, USA). Toutes les données obtenues ont été moyennées sur une durée d'enregistrement continue durant entre 5 et 10 minutes.In each protocol, a daily clinical examination of the mice was carried out to assess the viability of the mouse and measure its weight. Continuous non-invasive recording of transcutaneous oximetry, heart rate and respiratory rate was carried out on vigilant mice daily using the MouseOx® Plus device (Starr Life Sciences Corp, USA). All data obtained were averaged over a continuous recording duration lasting between 5 and 10 minutes.
1.81.8 Prélèvement biologiques et bio-conservationBiological sampling and bio-preservation
Immédiatement après le sacrifice de la souris, des prélèvements sanguins et des lavages broncho-alvéolaires (LBA) ont été réalisés et les poumons ont été prélevées. Les prélèvements sanguins ont été réalisés par section des gros vaisseaux thoraciques et stockés dans des aliquotes placés à température ambiante jusqu'à coagulation complète. Au décours, les aliquotes de sang coagulés ont été centrifugés à 1500g pendant 5 minutes à température ambiante. Le sérum et le culot de centrifugation ont alors été séparés et le sérum a été conservé à -20°C. Les LBA ont été réalisés uniquement sur le poumon droit après ligature soigneuse de la bronche souche du poumon gauche, par deux lavages successifs de 500 µL de PBS stérile, puis stockés immédiatement dans des aliquots placés dans de la glace. Les LBA ont alors été centrifugés à 1500g pendant 10 minutes à 4°C. Le surnageant et le culot de centrifugation ont été séparés et le surnageant conservé à -20°C. Le culot de centrifugation a été suspendu à nouveau dans 1 mL de PBS et homogénéisé. Les cellules re-suspendues ont été comptées manuellement sur cellule de Thoma. 100 000 cellules ont alors été étalées sur une lame de verre grâce à un Shandon Cytospin 4 (Thermo electron corporation, USA), puis colorées par une coloration de May-Grünwald Giemsa. Le poumon gauche a été placé dans une solution d'Antigenfix® (réf. 2545826, MM FRANCE, France) pendant 4 heures, rincé par deux bains successifs de PBS, puis progressivement déshydraté par immersion successive dans des bains d’alcool ayant une concentration croissante. Les pièces de poumons ont ensuite été placées dans un bain de Diasolv® (réf. 2545839, MM FRANCE, France) pendant 30 minutes puis inclues en paraffine pendant toute une nuit.Immediately after mouse sacrifice, blood samples and bronchoalveolar washings (BAL) were performed and lungs were removed. Blood samples were taken by sectioning the large thoracic vessels and stored in aliquots placed at room temperature until complete coagulation. Subsequently, the aliquots of clotted blood were centrifuged at 1500 g for 5 minutes at room temperature. The serum and the pellet were then separated and the serum was stored at -20°C. BALs were performed only on the right lung after careful ligation of the main bronchus of the left lung, by two successive washes of 500 µL of sterile PBS, then stored immediately in aliquots placed on ice. The BALs were then centrifuged at 1500 g for 10 minutes at 4°C. The supernatant and the centrifugation pellet were separated and the supernatant stored at -20°C. The centrifugation pellet was resuspended in 1 mL of PBS and homogenized. The resuspended cells were counted manually on a Thoma cell. 100,000 cells were then spread on a glass slide using a Shandon Cytospin 4 (Thermo electron corporation, USA), then stained with a May-Grünwald Giemsa stain. The left lung was placed in a solution of Antigenfix® (ref. 2545826, MM FRANCE, France) for 4 hours, rinsed with two successive baths of PBS, then gradually dehydrated by successive immersion in alcohol baths having a concentration increasing. The lung pieces were then placed in a Diasolv® bath (ref. 2545839, MM FRANCE, France) for 30 minutes then embedded in paraffin overnight.
1.91.9 Comptage des cellules inflammatoire et examen histologiqueInflammatory cell counting and histological examination
Pour chaque spot cellulaire de LBA préparé par Cytospin, un minimum de 200 cellules ont été manuellement comptées par un opérateur en aveugle du groupe expérimental de la souris étudiée. Chaque leucocyte visualisé a été catégorisé en tant que polynucléaire neutrophile, macrophage/monocyte, ou lymphocyte. Les proportions respectives et les nombres absolus de chaque type cellulaire ont été calculés. Les poumons inclus en paraffine ont été préparés pour l’analyse histologique à l’aide d’un microtome de façon à obtenir des coupes de 5 µm d’épaisseur. Les lames ont ensuite été colorées et examinées par un anatomopathologiste entrainé et en aveugle du groupe expérimental de la souris, de manière à déterminer le score Lung Injury Scoring System (LISS) comme décrit par [10].For each BAL cell spot prepared by Cytospin, a minimum of 200 cells were manually counted by a blinded operator from the experimental group of the mouse studied. Each visualized leukocyte was categorized as a neutrophil, macrophage/monocyte, or lymphocyte. The respective proportions and absolute numbers of each cell type were calculated. The paraffin-embedded lungs were prepared for histological analysis using a microtome to obtain 5 µm thick sections. The slides were then stained and examined by a trained pathologist blinded to the mouse experimental group, to determine the Lung Injury Scoring System (LISS) score as described by [10].

La concentration des LBA en mMPO a été déterminée par le kit mMPO DuoSet ELISA kit (réf. DY3667, R&D Systems, USA). La concentration des LBA en mTNFα a été déterminée par le kit mTNFα DuoSet ELISA (réf. DY410, R&D Systems, USA). Le dosage des taux sériques en endocan a été réalisé par le kit DIYEK M1 ELISA kit (réf. LIK-1101, Biothelis, France).The serum and BAL samples were thawed and then homogenized by shaking. The protein concentration of BAL was determined by the Pierce™ BCA Protein Assay kit (cat. no. 23227, Thermo Fisher Scientific, USA). The concentration of BAL in mMPO was determined by the mMPO DuoSet ELISA kit (ref. DY3667, R&D Systems, USA). The mTNFα concentration of BAL was determined using the mTNFα DuoSet ELISA kit (ref. DY410, R&D Systems, USA). The determination of serum levels in endocan was carried out using the DIYEK M1 ELISA kit (ref. LIK-1101, Biothelis, France).
1.111.11 StatistiquesStatistics
Toutes les analyses statistiques ont été réalisé avec le logiciel SPPS® 26 software (IBM, USA) pour Windows et tous les graphiques présentés avec le logiciel GraphPad Prism 6.0 software. La normalité des variables quantitatives n’a pas été retenue du fait des faibles effectifs. Un test de Mann–Whitney a été utilisé pour comparer la distribution des variables quantitatives non appariées. Pour le premier protocole, l’évaluation de l’effet du groupe expérimental de chaque souris sur les paramètres inflammatoires et l’évolution de l’oxymétrie, a été testé à l’aide d’un modèle mixe linéaire sur l’ensemble de la période étudiée de J0 à J10 en utilisant une approximation de Satterwaite. La variable « Groupe » a été considérée comme un effet fixe et la variable « Temps » comme un effet variable. Nous avons évalué l’effet du groupe expérimental de la même manière pour l’évolution de l’oxymétrie dans le second protocole. Les distributions des évaluations répétées dans le temps d’une même variable quantitative ont été testées par un test de Kruskal-Wallis. Tous les tests ont été réalisés en bilatéral avec un risque alpha fixé à 0.05.All statistical analyzes were carried out with SPPS® 26 software (IBM, USA) for Windows and all graphs presented with GraphPad Prism 6.0 software. The normality of the quantitative variables was not retained due to the low numbers. A Mann–Whitney test was used to compare the distribution of unpaired quantitative variables. For the first protocol, the evaluation of the effect of the experimental group of each mouse on the inflammatory parameters and the evolution of oximetry, was tested using a linear mixed model over the entire period studied from D0 to D10 using a Satterwaite approximation. The “Group” variable was considered as a fixed effect and the “Time” variable as a variable effect. We evaluated the effect of the experimental group in the same way for the evolution of oximetry in the second protocol. The distributions of repeated evaluations over time of the same quantitative variable were tested using a Kruskal-Wallis test. All tests were carried out bilaterally with an alpha risk set at 0.05.
2.2. RésultatsResults
2.12.1 L’endocan endogène murin diminue la sévérité de l’inflammation aigue pulmonaire et améliore la dysfonction physiologique pulmonaire.Murine endogenous endocan reduces the severity of acute pulmonary inflammation and improves pulmonary physiological dysfunction.
Dans notre premier modèle d’inflammation aigue pulmonaire induite par le LPS, nous avons mis en évidence que le taux sérique d’endocan variait en fonction du temps (p < 0.001) (Figure 2A). Les taux sériques médian d’endocan murin étaient de 2.59 ng.mL-1[2.39; 2.78] à J0, 0.42 [0.35; 0.68] à H12 et 2.28 [1.94; 2.57] à J10.In our first model of acute pulmonary inflammation induced by LPS, we demonstrated that the serum endocan level varied over time (p < 0.001) (Figure 2A). The maximum leukocyte recruitment rate was on D3 for WT mice and D4 for Esm1 -/- mice.
Sur la même période, des résultats comparables étaient trouvés pour les polynucléaires neutrophiles avec une diminution du nombre de neutrophiles retrouvés chez les souris WT versus les souris Esm1-/-(p < 0.001) (Figure 2C). Le recrutement alvéolaire en neutrophile était moins important chez les souris WT versus Esm1-/-à J1, J2, J4 et J5 (respectivement à J1: 335x103cellules [278; 406] vs 696 x103cellules [602; 1425] p = 0.003, à J2: 853x103cellules [689; 950] vs 1117x103cellules [1029; 1756] p = 0.007, à J4: 577x103cellules [195; 1765] vs 1932x103cellules [1251; 2153] p = 0.02 et à J5: 365x103cellules [198; 646] vs 1636x103cellules [1407; 1698] p = 0.01) (Figure 2C). Maximum recruitment of neutrophils was observed on D3 for WT mice versus D4 for Esm1 -/- mice.
Sur l’ensemble de la période, nous n’avons pas mis en évidence de différence entre les groupes concernant le recrutement de macrophage (p = 0.57) (Figure 2D). Le recrutement maximal en macrophage était observé à J5 chez les souris WT (687x103cellules.mL-1[620; 940]) contre J6 chez les souris Esm1-/-(536x103cellules.mL-1[358; 753]).Over the entire period, we did not demonstrate any difference between the groups regarding macrophage recruitment (p = 0.57) (Figure 2D). Maximum macrophage recruitment was observed on D5 in WT mice (687x10 3 cells.mL -1 [620; 940]) compared to D6 in Esm1 -/- mice (536x10 3 cells.mL -1 [358; 753]) .
Sur la période allant de J0 à J10, les concentrations de mMPO dans les LBA étaient moins importante chez les souris WT que chez les souris Esm1-/-(p < 0.001) (Figure 2E). A J5 et J6, les concentrations de mMPO dans les LBA étaient plus faible chez les souris WT comparées aux souris Esm1-/-(respectivement 547 µg.mL-1[441; 1110] vs 1437 µg.mL-1[1200; 1657] p = 0.015, et 211 µg.mL-1[191; 417] vs 796 µg.mL-1[512; 1070] p < 0.001). Le second protocole a examiné les effets biologiques d’une administration continue et sous cutanée d’endocan recombinant murin, 5 jours après l’administration intratrachéale de LPS. Au moment de l’administration intratrachéale de LPS (D0), les taux sériques d’endocan observés chez les souris Esm1-/-traitées par endocan murin, étaient de 67.8 ng.mL-1[45.9; 85.3].The second protocol examined the biological effects of continuous, subcutaneous administration of murine recombinant endocan, 5 days after intratracheal administration of LPS. 3.3. DiscussionDiscussion
Les tableaux de SDRA sont des entités syndromiques graves secondaires à une atteinte alvéolaire diffuse (DAD). Cette atteinte alvéolaire peut être la conséquence d’un recrutement non contrôlé des polynucléaires neutrophiles du compartiment sanguin vers le compartiment alvéolaire. (2) L’endocan, un protéoglycane excrété par l’endothélium pulmonaire surexprimé en condition inflammatoire, pourrait, en se liant à l’intégrine leucocytaire LFA-1 entrainant une limitation des mécanismes de diapédèse leucocytaire, avoir un rôle anti-migratoire sur les neutrophiles. [11]ARDS tables are serious syndromic entities secondary to diffuse alveolar damage (DAD). This alveolar damage may be the consequence of uncontrolled recruitment of neutrophils from the blood compartment to the alveolar compartment. (2) Endocan, a proteoglycan excreted by the pulmonary endothelium overexpressed in inflammatory conditions, could, by binding to the leukocyte integrin LFA-1 leading to a limitation of leukocyte diapedesis mechanisms, have an anti-migratory role on neutrophils. [11]
Pour permettre de pouvoir comparer nos résultats avec les données déjà disponibles, nous avons recueilli tous les paramètres inflammatoires ainsi que réalisé les examens histologiques selon les recommandations internationales de l’American Thoracic Society traitant des mesures validées pour l’étude des inflammations aigues pulmonaires dans des modèle animaux [10]). Pour étudier spécifiquement les effets de l’endocan, toutes les souris C57Bl/6 utilisées dans cette étude étaient identiques en terme d’âge, de poids et de conditions d’élevage. La seule condition qui différait était la présence ou non d’endocan. Pour étudier de manière spécifique les effets d’une administration continue d’endocan, nous avons choisi d’avoir recours à des osmopompes sous cutanées permettant une administration de traitement continue et à débit constant sur une durée totale de 3 jours. Nous avons ensuite sacrifié les souris à J5, au regard des résultats acquis lors du premier protocole réalisé. Pour l’administration d’endocan, nous avons eu recours uniquement à la forme glycosylée de l’endocan murin. Pour s’assurer de la pureté de la forme glycosylée et de l’absence de contaminant, des étapes de qualification ont été réalisé de manière systématique comme décrit précédemment.To enable us to compare our results with the data already available, we collected all the inflammatory parameters as well as carried out the histological examinations according to the international recommendations of the American Thoracic Society dealing with validated measurements for the study of acute pulmonary inflammations in animal model [10]). To specifically study the effects of endocan, all C57Bl/6 mice used in this study were identical in terms of age, weight and rearing conditions. The only condition that differed was the presence or absence of endocan. To specifically study the effects of continuous administration of endocan, we chose to use subcutaneous osmopumps allowing continuous administration of treatment at a constant flow rate over a total duration of 3 days. We then sacrificed the mice on D5, based on the results acquired during the first protocol carried out. For the administration of endocan, we used only the glycosylated form of murine endocan. To ensure the purity of the glycosylated form and the absence of contaminants, qualification steps were carried out systematically as described previously.
Les résultats du premier protocole ont mis en évidence que la présence d’endocan endogène (souris WT) diminue la sévérité de l’atteinte inflammatoire pulmonaire induite par le LPS, et améliore la dysfonction physiologique pulmonaire (SpO2supérieur dans le groupe WT) (
Dans notre second protocole, l’administration continue d’endocan glycosylé murin chez des souris Esm1-/-au décours d’une instillation intra-trachéale de LPS, a d’une part diminué les paramètres de l’inflammation pulmonaire et d’autre part amélioré la dysfonction physiologique pulmonaire. Nous avons vérifié par des dosages sériques que lors de l’administration continue d’endocan il existait bien de l’endocan circulant. Les résultats montrent des taux circulants d’endocan murin glycosylé se situant entre 60 et 70 ng/ml. Sur l’ensemble de la période entre J0 et J5, le traitement par endocan a entrainé une diminution significative du nombre de leucocytes, du nombre de neutrophiles et de macrophages recrutés au niveau alvéolaire, mais aussi a entrainé une amélioration des paramètres de l’oxygénation sanguine (
Au total, toutes ces valeurs confirment que la présence d’endocan (endogène ou exogène) peut, en réponse à un stimulus inflammatoire, non seulement diminuer l’importance de l’inflammation aigue pulmonaire mais aussi améliorer la défaillance physiologique pulmonaire. Par ailleurs, nous avons observé un effet dose avec une réponse plus importante si la concentration sérique en endocan dépasse les taux physiologiques (second protocole).In total, all these values confirm that the presence of endocan (endogenous or exogenous) can, in response to an inflammatory stimulus, not only reduce the importance of acute pulmonary inflammation but also improve physiological pulmonary failure. Furthermore, we observed a dose effect with a greater response if the serum concentration of endocan exceeds physiological levels (second protocol).

Nous avons étudié l’effet de la forme endogène de l’endocan ainsi que l’effet d’une administration continue d’endocan permettant d’obtenir des concentrations sériques supra physiologiques. Des expérimentations préliminaires avaient montré que des taux sériques élevés en endocan ne pouvaient être atteint et maintenus pendant plusieurs heures au décours d’une injection intra-péritonéale unique en endocan (données non publiées). De façon à mieux étudier l’effet de l’administration d’endocan, une administration par perfusion continue semble donc à privilégier.We studied the effect of the endogenous form of endocan as well as the effect of continuous administration of endocan to obtain supraphysiological serum concentrations. Preliminary experiments had shown that high serum levels of endocan could not be achieved and maintained for several hours following a single intraperitoneal injection of endocan (unpublished data). En conclusion, les résultats ont montré qu’au décours d’un stimulus inflammatoire aigu pulmonaire par instillation intra-trachéale de LPS chez la souris C56Bl/6, la perfusion continue d’endocan chez des souris Esm1-/-permet de diminuer significativement l’inflammation pulmonaire et améliorer la dysfonction physiologique pulmonaire.In conclusion, the results showed that following an acute pulmonary inflammatory stimulus by intratracheal instillation of LPS in C56Bl/6 mice, the continuous infusion of endocan in Esm1 -/- mice significantly reduces the pulmonary inflammation and improve pulmonary physiological dysfunction.
Ces résultats confirment que l’endocan glycosylé constitue une solution thérapeutique efficace pour traiter les SDRA.These results confirm that glycosylated endocan constitutes an effective therapeutic solution for treating ARDS.
Claims (13)
