La présente demande de brevet concerne un procédé d’extraction de composés hydrosolubles d’intérêt nutritionnels à partir d’une culture de microalgues vivantes grâce à une perméation temporaire de la paroi cellulaire.This patent application concerns a process for extracting water-soluble compounds of nutritional interest from a culture of living microalgae using temporary permeation of the cell wall.
En raison des faibles volumes de production des microalgues dans le monde et de leur coût élevé, leur utilisation reste encore très limitée notamment dans le domaine de l’alimentation humaine.Due to the low production volumes of microalgae worldwide and their high cost, their use still remains very limited, particularly in the field of human food.
De nombreux industriels préfèrent ainsi valoriser les molécules à haute valeur ajoutée telle de la phycocyanine issue de la spiruline, ou bien l’astaxanthine extraite d’Haematococcus pluvialisdestinées à des applications pharmaceutiques.Many manufacturers thus prefer to promote molecules with high added value such as phycocyanin from spirulina, or astaxanthin extracted from Haematococcus pluvialis intended for pharmaceutical applications.
Le secteur de l’alimentaire quant à lui est confronté à de nombreux obstacles dont l’organoleptique constitue le principal. En effet, malgré leurs propriétés nutritionnelles exceptionnelles, les microalgues restent très peu consommées telles quelles du fait de leurs caractéristiques organoleptiques quelque peu rebutantes pour le consommateur lambda.The food sector is faced with numerous obstacles, the main one being organoleptics. Indeed, despite their exceptional nutritional properties, microalgae are rarely consumed as they are due to their organoleptic characteristics that are somewhat off-putting for the average consumer.
Pour pallier ce problème, de nombreux acteurs recourent à des procédés d’extraction et de fractionnement de la biomasse afin d’éliminer les contraintes organoleptiques et de ne conserver que les éléments d’intérêt nutritionnel/santé.To overcome this problem, many players use biomass extraction and fractionation processes in order to eliminate organoleptic constraints and only retain elements of nutritional/health interest.
Ainsi Soto-Sierra (2018) décrit différentes méthodes d’extraction physiques permettant d’extraire les protéines des algues pour des applications alimentaires comme les ultrasons, le broyage à billes, l’homogénéisation haute pression. Toutes ces techniques permettent l’obtention d’un extrait riche en protéines, séparé des parois cellulaires vertes, qui est ensuite purifié puis séché avant utilisation. Ces techniques peuvent être appliquées à diverses espèces de microalgues comme la spiruline, la chlorelle et bien d’autres. De manière similaire, Bleakley (2017) présente les techniques d’extraction appliquées aux algues pour en extraire les protéines destinées à des usages alimentaires pour l’homme. On peut ainsi citer la méthode biologique faisant intervenir l’hydrolyse enzymatique, le recourt à des procédés physiques ou bien les méthodes chimiques avec des traitements acides par exemple.Soto-Sierra (2018) describes different physical extraction methods for extracting proteins from algae for food applications such as ultrasound, ball milling, high pressure homogenization. Ainsi, le brevet US2012095245 (A1) (2012) mentionne l’emploi combiné d’un traitement aux ultrasons et l’usage d’un solvant organique pour extraire les lipides deNannochloropsistout en maintenant la culture vivante après traitement. De manière similaire, Hejazi (2004) recourt à un système de réacteur à deux phases permettant le passage de dodécane à contre-courant de la culture deDunaliella salinapour l’extraction en continue du bêta-carotène. Durant la période d’extraction, le dodécane, qui est un solvant biocompatible avec les algues, est renouvelé pour une meilleure efficacité d’extraction. Une fois, les cellules traites, elles sont remises en culture. De la même façon, Samori (2019) emploie du cyclohexane pour l’extraction biocompatible de l’astaxanthine issue de cultures vivantes d’Haematococcus pluvialis.In order to reduce the number of unit operations and continue to work with the same renewed biomass, certain players have become interested in so-called non-destructive extraction techniques. Ces techniques biocompatibles présentent l’avantage de réduire le nombre d’opérations unitaires et de renouveler la culture d’algues contrairement à un procédé classique d’extraction. Néanmoins, ces méthodes nécessitent l’usage en grande quantité de solvants organiques peu écologiques et souvent interdits en alimentaire.These biocompatible techniques have the advantage of reducing the number of unit operations and renewing the cultivation of algae unlike a traditional extraction process. Le Demandeur a résolu ce problème technique en mettant au point une technique permettant l’extraction de composés d’intérêt nutritionnels à partir d’une culture d’algues vivantes sans recours à des solvants organiques. Une perméation temporaire de la paroi induite par des champs électriques pulsés permet ainsi la sortie des composés d’intérêt de la cellule et leur solubilisation dans le milieu d’extraction. Les cellules de microalgue ayant été traitées par champs électriques pulsés sont ensuite remises en culture pour une future utilisation permettant ainsi de réduire considérablement le coût du procédé global ainsi que le nombre d’opérations unitaires. En effet, avec cette technique, il n’est pas nécessaire de récolter ni de sécher la biomasse avant extraction.The Applicant has solved this technical problem by developing a technique allowing the extraction of compounds of nutritional interest from a culture of living algae without the use of organic solvents. Ainsi, selon un premier aspect, l’invention concerne un procédé d’extraction de composés hydrosolubles d’intérêt nutritionnels, de préférence des protéines, vitamines, minéraux, ou sucres, à partir d’une culture de microalgues vivantes comprenant les étapes de :
- Mise en culture des souches de microalgues jusqu’à obtenir une quantité de biomasse desdites microalgues de concentration comprise entre 1. 106et 1.108cellules/mL ;
- Séparation des microalgues obtenues à l’étape a. de leur milieu de culture, de préférence par centrifugation entre 1500 et 4000 g durant 5 à 20 minutes et à une température comprise entre 4 et 25°C ;
- Mélange des microalgues obtenues à l’étape b. dans de l’eau à un ratio compris entre 5 et 20% à une conductivité comprise entre 20 et 200 µS/cm ;
- Application au mélange obtenu à l’étape c. de champs électriques pulsés bipolaires, de préférence au nombre de 1 à 20 impulsions avec une durée d’impulsion de 1 à 2 ms, un voltage compris entre 0,5 et 6 kV/cm, une température comprise entre 4 et 25°C, et un débit de circulation des cellules algales dans la chambre d’impulsion de 10 à 100 mL/min,
- Dilution du mélange obtenu à l’étape d. entre 2 et 5 fois dans de l’eau ou un tampon durant entre 1 et 12h ;

- Mélange des microalgues obtenues à l’étape b. dans de l’eau à un ratio compris entre 5 et 20% à une conductivité comprise entre 20 et 200 µS/cm ;
- Application au mélange obtenu à l’étape c. de champs électriques pulsés bipolaires, de préférence au nombre de 1 à 20 impulsions avec une durée d’impulsion de 1 à 2 ms, un voltage compris entre 0,5 et 6 kV/cm, une température comprise entre 4 et 25°C, et un débit de circulation des cellules algales dans la chambre d’impulsion de 10 à 100 mL/min,
- Dilution du mélange obtenu à l’étape d. entre 2 et 5 fois dans de l’eau ou un tampon durant entre 1 et 12h ;
- Centrifugation du mélange obtenu à l’étape e., de préférence à entre 1500 et 4000 g durant 5 à 20 minutes à une température comprise entre 4 et 25°C ;
- Récupération du surnageant de centrifugation comprenant lesdits composés d’intérêt nutritionnel ;
- Obtention de composés d’intérêt nutritionnel.
