Description
The present invention relates to the field of natural extracts as cosmetic active ingredients. The subject of the invention is in particular an extract of oak buds of the Quercus genus, its process for obtaining it and its cosmetic uses.
Etat de l’artState of the art
The appearance of the face, neck and décolleté is considered by many women both as a symbol of femininity and as an important indicator of age.
As we age, the appearance of the skin naturally deteriorates and reveals sagging skin, sagging of the skin on the cheeks and neck, thinner, drier and more sensitive skin as well as more pronounced wrinkles.
When the skin presents these characteristics linked to aging, we then speak of mature skin. Mature skin has its own characteristics. Indeed, with age and exposure to ultraviolet (UV), the skin is damaged, spots appear and the layers of the skin become disorganized.
In addition, within mature skin, cell renewal and microcirculation are slowed down. The complexion becomes dull, loses uniformity and the skin is more prone to redness. However, the radiance of the complexion reflects the health of the skin, a dull complexion makes you look tired and can accentuate the signs of aging.
Mature skin needs good hydration, allowing it to restructure and redensify the skin and improve the radiance of the complexion. However, finer and more sensitive, it requires a non-aggressive routine.
To meet this need, the cosmetics industry is particularly interested in natural extracts with beneficial effects for the skin. Indeed, consumers of cosmetic products are increasingly sensitive to the use of natural extracts, such as products from gemmotherapy.
Gemmotherapy is a subfamily of herbal medicine aimed at using the buds of trees and plants. Gemmotherapy is based in particular on the use of young and fresh plant material. Extracts from buds are mainly used as a food supplement for different properties which depend on the origin of the plant or tree, such as detoxifying, slimming, cholesterol-fighting properties, etc.
Some plant bud extracts are already known for their cosmetic use, for example beech bud extracts.
However, the cosmetic industry and consumers are always looking for new natural extracts with improved anti-aging effects, more particularly extracts with effective anti-aging action, particularly on mature skin.
To meet this need, the invention proposes a new extract obtained from buds of the Quercus genus comprising in particular polyphenols and sugars.
Extracts of Fagaceae are already known, for example, patent application FR1870215 describes extracts of bast and/or cambium for cosmetic uses.
The cambium is a thin layer of meristematic cells located between the wood (secondary xylem) and the bast. The bast, or secondary phloem, is made up of sieve tubes, their companion cells, parenchyma and fiber. It is produced by the cambium towards the exterior. The Liber and the Cambium are an integral part of the trunk of the tree and are very distinct from the buds.
Thus, no extract of buds of the Quercus genus has been described for anti-aging cosmetic effects.
However, the inventors surprisingly discovered that an extract of oak buds of the Quercus genus made it possible to obtain such an anti-aging effect. In particular, said extract makes it possible to redensify, restructure or even revitalize the skin.
The extract according to the invention is preferably obtained from oak buds of the species Quercus petraea and/or Quercus robur.
The oak of the Quercus petraea species, also called sessile oak, is a species of tree in European forests with a subatlantic tendency. This species of oak corresponds to trees that can reach up to 40 meters in height, with deciduous foliage and presenting buds with a pointed ovoid shape.
The oak of the Quercus robur species, also called pedunculate oak, is a sub-oceanic Eurasian tree species.
By “extract” within the meaning of the invention, we mean a set of molecules extracted from a raw material X, native or resulting from transformation (for example by digestion). Thus, by “oak extract of the Quercus genus”, in the sense of the invention, is meant a set of several molecules obtained by an action, on the plant raw material, in this case oak of the Quercus genus, in particular buds of the oak genus Quercus , more preferably of the species Quercus petraea and/or Quercus robur . Preferably, the action making it possible to obtain this set of molecules is an extraction, more particularly a digestion of the buds of the oak of the genus Quercus .
By “digestion” within the meaning of the invention, is meant the process of extracting all of the molecules extracted from a raw material X, in particular the oak bud of the genus Quercus . Said digestion is carried out by heating in a liquid below the boiling point. Preferably, the liquid is chosen from water and a mixture of water/glycerin. Pure glycerin has a boiling point of 290°C. Those skilled in the art adapt the heating temperature according to the proportion of water/glycerin of the mixture used for digestion.
By “buds” within the meaning of the invention, we mean a set of very young leaf or floral pieces grouped on a very short axis surrounding a growing point. It generally corresponds to an outgrowth appearing on certain parts of plants and giving rise to branches, leaves, flowers and fruits.
By “cosmetic active ingredient” within the meaning of the invention is meant a set of molecules having non-therapeutic cosmetic effectiveness on healthy skin, in particular an anti-aging effect.
By “mature skin” within the meaning of the invention, we mean a condition of the skin from a certain stage of skin aging. This natural process concerns all skin types (oily, dry). Mature skin is characterized by skin that sags and shows a loss of volume in different areas of the face (corners of the lips, eyes) or neckline. On the surface, this results in the appearance of wrinkles, but also in skin that appears thinner, more fragile, drier and less toned.
By “reinforcing the barrier function of the skin” within the meaning of the invention, we mean reinforcing the cohesion of the upper layers of the epidermis, in particular by stimulating the formation of tight junctions or by reducing the desquamation of the stratum corneum. The strengthening of the barrier function of the skin can be measured by quantifying the expression of filaggrin, a structural protein and marker of terminal differentiation of the epidermis.
By “regeneration of the dermis” within the meaning of the invention is meant the stimulation of the synthesis of dermis proteins such as lumican, which structures the dermis.
By “promoting the organization of the extracellular matrix” within the meaning of the invention is meant the stimulation of the expression of dermatopontin which regulates cell-matrix interactions and the assembly of the matrix.

The subject of the present invention is therefore an extract obtained from oak buds of the Quercus genus comprising polyphenols and sugars.
The raw material from which the extract originates can be any tree of the genus Q u ercus , and in particular chosen from the following species: petraea, robur, alba, rubra, cerris, pubescen s , pyrenaica, ilex, suber, coccifera, and variabilis . Preferably, it is Quercus petraea and/or Quercus robur .
Also, the extract according to the invention is obtained preferentially from oak buds of the species Quercus petraea and/or Quercus robur , even more preferentially from oak buds of the species Quercus petraea and/or Quercus robur from the Bertranges forest in France.

According to a preferred object, the extract according to the invention comprises a polyphenol content of at least 2g/L gallic acid equivalent, measured by the Folin-Ciocalteu method. The Folin-Ciocalteu method is a method well known to those skilled in the art for measuring the total polyphenol content. The total polyphenol content is expressed in gallic acid equivalent which is proportional to the quantity of polyphenols present in the extract. The gallic acid equivalent is calculated in particular by producing a standard range of the absorbance of gallic acid in relation to its concentration. The equation obtained from the standard range makes it possible to precisely measure the gallic acid content in the extract according to the invention.

Preferably, the extract according to the invention also has an anti-radical activity of at least 1g/L quercetin equivalent. The anti-radical activity can be measured by a DPPH test, a technique well known to those skilled in the art. Thus, the oak buds obtained in step a) are preferentially frozen before carrying out step b). Also, the extract according to the invention can be obtained from frozen buds of oak of the genus Quercus , preferably of the species Quercus petraea , followed by a drying step, a step of grinding the dried buds and digestion of the ground material obtained in the previous step with an extraction solvent chosen from water, and a mixture of water and glycerin, at a temperature between 40 and 70°C.
Thus, according to a particularly preferred embodiment, the extract according to the invention can be a hydro-glycerin extract or an aqueous extract, preferably a hydro-glycerin extract. After extraction, the extract is generally brown in color but can be discolored and/or recolored by any techniques known to those skilled in the art.

- Harvesting oak buds of the genus Quercus ,
- Drying of the buds obtained in step a) at a temperature between 40 and 120°C.

De façon particulièrement préféré, l’extrait selon l’invention est capable d’augmenter l’expression de la dermatopontine, du lumican, de la filaggrine ou de la protéine ZAG.Particularly preferably, the extract according to the invention is capable of increasing the expression of dermatopontin, lumican, filaggrin or the ZAG protein.
L’augmentation de son expression favorise ainsi l’organisation et l’architecture de la peauDermatopontin is an extracellular matrix protein highly expressed in the dermis whose functions are cell-matrix interactions and matrix assembly. The increase in its expression thus promotes the organization and architecture of the skin
Lumican is an extracellular matrix protein involved in the organization and growth of collagen fibers as well as epithelial cell migration and tissue repair. The increase in its expression thus promotes the organization of the extracellular matrix, the restructuring of the layers of the skin, and therefore the regeneration of the dermis.
Filaggrin is a structural protein that is fundamental in the development and strengthening of the epidermal barrier. It is a marker of terminal differentiation of the epidermis. In addition, the production of natural hydration factors results from the digestion of this protein. Filaggrin is associated with intermediate keratin filaments and aggregates them within the epidermis.
