Description
The invention proposes a process for diluting or mixing fluids injected sequentially under a controlled atmosphere.
Selon l’invention, le procédé comprend:According to the invention, the method comprises:
An injection of a controlled volume of at least one liquid inside at least one chamber by means of at least one pressure controller coupled to at least one selector valve, said injection is done by at least one liquid inlet into said at least one chamber.
At least one reactor comprising said at least one chamber with at least one inlet for fluids or at least two inlets, at least one for liquids and at least one for gases. Said at least one chamber can be pressurized and the liquids in said at least one chamber agitated with at least one rotating magnetic object to allow dilution, mixing or chemical or biochemical reaction.
A pressure increase achieved by the introduction of fluids through said at least one inlet into said at least one reactor until a controlled and defined pressure less than or equal to 10 bar is reached, said pressure producing a displacement at a controlled rate of the volume of liquid present in the reactor by at least one fluid outlet regulated by at least one valve connecting said volume of liquid and the outside of said reactor.
A pressure reduction in said at least one reactor until reaching a controlled and defined pressure less than or equal to 10 bar, said pressure reduction accomplished by at least one vent valve and allowing the introduction of a new fluid without producing displacement of the volume present in the reactor by said at least one fluid outlet connecting said volume of liquid and the outside of said reactor.
The method according to the invention makes it possible to create a system based on the sequential injection of liquids into a potentially pressurized secondary reactor. This system is used not only to make mixtures of liquids but also to move a fluid, create controlled dilutions of a given substance, carry out chemical reactions under controlled atmosphere and enrich the mixture with gases using the principle of magnetic stirring in a pressurized tank.
Advantageously, the method according to the invention makes it possible to carry out chemical and biochemical reactions involving finite volumes of reagents under controlled atmosphere and pressure in a completely isolated environment. In the invention, many gases can be used such as, for example, nitrogen, oxygen or carbon dioxide.
Alternatively, a volume of liquid larger than the reactor can be pushed entirely into the reactor sequentially or continuously using gas to enrich the liquid with gas.
The invention also relates to a device implementing the method described above, where said displacement of said volume of liquid by said at least one fluid outlet connecting said volume and the outside of said at least one reactor takes place towards a microfluidic device where the flow becomes laminar again.
In a variant, the reaction takes place separately and in parallel with at least one main laminar flow of liquid. In this case, said at least one fluid outlet is towards said at least one main laminar flow of liquid.
The invention adapts to any type of gas and liquid and the thermalization of the reactor is possible if necessary.
The invention relates to the field of mixtures and dilutions of fluids for chemical and biochemical processes in microfluidic systems. In particular, the invention relates to the automated sequential injection of different reagents into a pressurized reaction chamber comprising a bar magnet. More particularly, the invention relates to the possibility of carrying out chemical reactions with precise volumes of reagents under a controlled atmosphere with the possibility of temperature control as well.
Many parameters including the order of the reagents, the mixing time, the temperature and the stoichiometry among others are controlled during the chemical reactions to increase the yield, the purity or the monodispersity of the product.
The use of microfluidics compared to conventional methods in chemistry and biochemistry, namely mixing and reaction in chambers connected and fed by microchannels characterized by fluidic volumes between 1 pL and 10 mL, makes it possible to increase the surface to volume ratio, which facilitates mass and heat transfer, thus increasing the yield and speed of reactions. Diagnoses and analyzes are therefore accelerated. Microfluidics also makes it possible to parallelize the tests and thus to massively increase the number of reactions. In addition, the volumes of reagents used are lower, making the processes greener, safer and reducing costs.
But despite the many advantages of microfluidics, continuous chemical and biochemical reactions are not yet fully optimized on a small scale. The chemical compatibility between solvents and materials is still a real challenge. It is also difficult to carry out reactions under a controlled atmosphere or to make liquid/gas or gas/gas mixtures. Problems related to the transport of unwanted bubbles in the microchannels are also common in some methods. In addition, the mixing and dilution of reagents remains a major challenge, the laminar flow present in microfluidic systems not facilitating this step. On the other hand, many reactions must be performed with a controlled sequential set of conditions, including the addition of minute reagents, which adds challenges when aiming to achieve microfluidic-scale reactions.
In microfluidics, two mixing techniques are used to improve the mixing of solutions (Lee C.-Y. et al, Microfluidic Mixing: A Review, Int J Mol Sci, 2011; 12(5): 3263-3287). On the one hand, passive mixing techniques (Lee C.-Y. et al. Passive mixers in microfluidic systems: A review. Chemical Engineering Journal, vol. 288, 2016, pages 146 to 160, ISSN 1385-8947) based on particular geometries of the microfluidic chips (T or Y junction, serpentine or 3D channels) make it possible to create turbulences in the flow and thus not to let only the diffusion mix the reagents. However, these passive techniques lack efficiency because uniform mixing of the solution is not achieved instantaneously. On the other hand, active mixing techniques improve mixing efficiency by using external forces (acoustic, magnetic or thermal). A uniform mixture is then instantly obtained.
Semenov describes in his publication a microreactor of internal volume 250 µL, activated by magnetic field, made of polydimethylsiloxane (PDMS) and glass, for the mixing of solutions (Semenov, S. et al, Nature, 537, 656–660, 2016 ). The microreactor has 3 liquid inlets for the injection of 3 different solutions and has an internal volume of 250 µL. The injection of fluids into the microreactor is controlled by syringe pumps. This system also has a thermostated channel for temperature control of the microreactor. This system therefore makes it possible to mix three different solutions in a homogeneous and instantaneous way and to carry out chemical reactions in a thermostatically controlled environment.
However, this system is not without drawbacks. The chemical reactions are limited to only 3 reactants, which drastically reduces the flexibility of the system. PDMS, a material widely used in microfluidics for the manufacture of chips, has limited resistance to chemical solutions and absorbs molecules, which modifies the stoichiometry of the reactions. In addition, this material is permeable to oxygen and chemical reactions under controlled atmosphere are therefore not possible. In addition, no gas enrichment of the solutions is possible. Finally, mixing at constant volume for several minutes is not possible. Finally, the system is not optimized to perform different chemical reactions in the same microreactor without risk of cross-contamination.
Patent application US6572255B2 (BECKMAN COULTER INC) describes a reactor and its fluidic system for mixing liquids in a controlled manner and delivering dilute solutions. Mixing is done by the recirculation method. The mixing chamber includes two liquid inlets in its upper part and an outlet and inlet in its lower part, allowing the installation of the recirculation loop. The chamber is equipped with ventilation protected by a filter to avoid any contamination of the environment. This chamber is equipped with a system for heating the solution as well as a temperature sensor to check the temperature of the mixture. In addition, a concentration sensor (here a conductometric sensor) is present to know the concentration of the mixture. Finally, a level sensor is integrated to avoid overflowing the chamber and having returns in the input circuit. The movement of the solution is controlled by a system of valves and pumps.
Despite all the advantages of this system, the fact of using a recirculation loop increases the dead volume present in the fluidic system. This method is therefore not optimal for working with small volumes of reagents or if uniform and instantaneous mixing is necessary. In addition, it is not possible to carry out the reaction under a controlled atmosphere, limiting the chemical reactions that can be carried out, the ventilation only working passively towards the outside. Finally, this system lacks flexibility because it is only possible to inject two solutions.
There is therefore a need for a reactor to carry out chemical and biochemical reactions in a system making it possible to sequentially inject reagents in a controlled and automated manner by reducing the volumes used. The chemical resistance of the materials in the system, as well as their inertness, is essential so as not to interfere in the reactions.
In addition, there is a need to carry out these reactions under controlled atmosphere or to have the possibility of mixing gases with liquids or gases together.
