Abstract
La présente invention concerne l’utilisation d’un agent antioxydant pour stabiliser le safran et d’une composition comprenant du safran ou un extrait de Crocus sativus associé à au moins un tel agent antioxydant. Une telle composition est particulièrement utile pour limiter la dégradation des molécules présentes dans le safran, en particulier le safranal et les crocines.
Description
L’invention concerne l’utilisation d’un agent antioxydant pour stabiliser le safran et d’une composition comprenant du safran ou un extrait deCrocus sativusassocié à au moins un tel agent antioxydant. Une telle composition est particulièrement utile pour limiter la dégradation des molécules présentes dans le safran, en particulier le safranal et les crocines.
Etat de l’art
Les extraits de safran connus dans l’art antérieur sont pour la plupart standardisés à 2% de safranal par spectrométrie UV selon la norme ISO 3632-2 :2010 qui surévalue la teneur en safranal de 20 à 50 fois (Garcia-Rodriguez et al., 2017 «Comparative evaluation of an ISO 3632 method and an HPLC-DAD method for safranal quantity determination in saffron»). C’est ainsi que de nouveaux extraits dont la teneur est mesurée par HPLC sont récemment apparus sur le marché, tels qu’un extrait commercialisé par la société Pharmactive titré à 0,03% (tel que décrit dans WO 2017/182688), mais encore par la société Activ’Inside titré à au moins 0,2% (tel que décrit dans FR 3054443).
Les extraits de safran sont bien connus pour avoir des propriétés bénéfiques pour l’Homme ou l’animal afin de lutter par exemple contre le stress, l’anxiété, la mauvaise humeur mais également certaines pathologies telles que la dépression ou les troubles cognitifs, par exemple la maladie d’Alzheimer.
Malgré leur efficacité, les extraits de safran connus et commercialisés souffrent d’instabilités importantes dans des conditions normales d’entrepôt et ce dès la fin du processus de fabrication, par exemple, lors du transport des marchandises ou lors du stockage des produits avant leur acheminement dans les différents points de vente.
Pour y remédier, ou au moins limiter cette instabilité, les produits peuvent être conservés à l’abri de la lumière, de l'humidité à hauteur de 40 à 60% et à une température comprise entre 20°C et 25°C.
Or, malgré des conditions normales de stockage, les extraits de safran restent instables compte tenu notamment des pertes en crocines et en safranal, deux molécules associées aux bienfaits du safran.
Il existe donc un besoin pour stabiliser les extraits de safran, limiter voir empêcher leur dégradation et ainsi protéger l’ensemble des molécules d’intérêt présentes dans le safran, en particulier les crocines et le safranal.
C’est l’objectif de la présente invention.
Pour y répondre, les inventeurs ont, de façon surprenante, observé que l’utilisation d’un agent antioxydant et en particulier son intégration dans une composition comprenant du safran ou au moins un extrait de safran permet de stabiliser des produits à base de safran ou d’extrait de safran et lutter contre la dégradation des crocines et du safranal.
Il a déjà été établi que des caroténoïdes dits hydrophobes (Beta-carotene, zeaxanthin, astaxanthin, et le lycopene) sont sensibles à l’oxydation notamment du fait de la présence d’un motif polyène conjugué, des formes époxy peuvent alors être générées lors d’une étape d’auto-oxydation. Cette oxydation peut alors être contrôlée par l’ajout d’un agent antioxydant (Yuwei Liu et al. 2015). Or, de tels caroténoïdes ne sont pas présents dans les stigmates deCrocus sativus.
Les crocines sont, pour leur part, sensibles à l’hydrolyse basique ou acide qui favorise leurs dégradations, les transformant alors en crocétine (CN Reddy et al. 2020). De plus, Moratalla-López et al. ont observé que la stabilité des crocines était également pH dépendante (Moratalla-López et al. 2018). Enfin, il est connu que la molécule de safranal est instable du fait de sa très grande volatilité et de son faible poids moléculaire.
Or, il est bien connu de l’homme de l’art que l’ajout d’un antioxydant pour empêcher l’hydrolyse, maintenir le pH ou réduire la volatilisation du safranal n’est pas d’intérêt.
D’autres études suggèrent l’utilisation de systèmes d’encapsulation de type émulsion conventionnelles ou multiples des caroténoïdes pour améliorer leur stabilité (CS Boon et al. 2010) mais également freiner la perte de force colorante (JC Cuevas-Bernardino et al. 2019), mesurée à travers l’absorbance aux trois longueurs d’onde décrites dans la méthode ISO3632 (440nm crocines, 330nm safranal, 257nm picrocrocine).
Toutefois, plusieurs travaux ont permis de démontrer que la force colorante mesurée par spectrométrie UV n’est pas corrélée avec la composition réelle en différentes crocines, picrocrocine et dérivés, safranal (B Moras et al. 2018). Ainsi, les systèmes décrits dans la littérature ne permettent pas de garantir la stabilité de l’ensemble des actifs du safran, en particulier les crocines et le safranal et donc de répondre au problème de l’invention.
Ainsi, l’invention concerne une composition comprenant au moins un stigmate deCrocus sativuset/ou au moins un extrait deCrocus sativuset au moins un agent antioxydant, le stigmate deCrocus sativuset/ou l’extrait deCrocus sativuscomprenant du safranal et des crocines, ledit agent antioxydant représentant entre 0,01% et 25% en poids par rapport au poids total de la matière sèche de crocines, mesuré par HPLC. Ainsi, le poids total de la matière sèche de crocines est mesuré par HPLC et l’agent antioxydant représente entre 0,01% et 25% en poids par rapport audit poids total de crocines.
Le ratio de l’agent antioxydant/safranal présent dans la composition selon l’invention est compris entre 1/100 et 10/1, en poids par rapport au poids total de la matière sèche de safranal, mesuré par HPLC.
Au moins un agent stabilisant et/ou chélateur peut également être ajouté dans la présente composition selon l’invention, celui-ci représente alors entre 0,01% et 25% en poids par rapport au poids total de la matière sèche de crocines, mesuré par HPLC.
Préférentiellement, le safranal compris dans le ou les stigmate(s) deCrocus sativuset/ou le ou les extrait(s) deCrocus sativus, représente au moins 0,03%, en poids par rapport au poids total de l’extrait deCrocus sativuset/ou stigmate deCrocus sativus, mesuré par méthode HPLC.
Les crocines présentent dans le ou les stigmate(s) deCrocus sativuset/ou le ou les extrait(s) deCrocus sativus, représentent préférentiellement au moins 1% de crocines, en poids par rapport au poids total de l’extrait deCrocus sativuset/ou stigmate deCrocus sativus, mesuré par méthode HPLC.
Préférentiellement, l’agent antioxydant présent dans la composition selon l’invention est choisi parmi l’acide ascorbique (E300), la vitamine E telle que l’α-tocophérol (E306) ou le δ-tocophérol (E309) ; un extrait de romarin E392, un extrait de mélisse, un extrait deVitis vinifera, un extrait de pétales deCrocus sativuscontenant des flavonols et plus particulièrement des kaempférols, le butylhydroxyanisol (BHA E320), le butylhydroxytoluène (BHT E321), et de l’acide citrique (E330).
Enfin, ladite composition selon l’invention peut être encapsulée ou microencapsulée.
Selon un autre aspect, l’invention se rapporte également à l’utilisation d’un agent antioxydant pour lutter contre la dégradation du safran (stigmate et/ou extrait deCrocus sativus) et/ou réduire son instabilité, préférentiellement du safranal et/ou des crocines.

- Composition selon la revendication précédente, caractérisée en ce que le ratio de l’agent antioxydant/safranal est compris entre 1/100 et 10/1, en poids par rapport au poids total de la matière sèche de safranal, mesuré par HPLC.
- Composition selon l’une des revendications précédentes, caractérisée en ce que la composition comprend en outre, un agent chélateur représentant entre 0,01% et 25% en poids par rapport au poids total de la matière sèche de crocines, mesuré par HPLC.
- Composition selon l’une des revendications précédentes, caractérisée en ce que la composition comprend au moins 0,03% de safranal, en poids par rapport au poids total de l’extrait deCrocus sativuset/ou stigmate deCrocus sativus, mesuré par méthode HPLC.
- Composition selon la revendication précédente, caractérisée en ce que le safranal représente au moins 0,1% en poids par rapport au poids total de l’extrait deCrocus sativuset/ou stigmate deCrocus sativus, mesuré par méthode HPLC.
- Composition selon la revendication précédente, caractérisée en ce que le safranal représente au moins 0,2% en poids par rapport au poids total de l’extrait deCrocus sativuset/ou stigmate deCrocus sativus, mesuré par méthode HPLC.
- Composition selon l’une des revendications précédentes, caractérisée en ce que la composition comprend au moins 1% de crocines, en poids par rapport au poids total de l’extrait deCrocus sativuset/ou stigmate deCrocus sativus, mesuré par méthode HPLC.
- Composition selon l’une des revendications précédentes, caractérisée en ce que la composition comprend au moins 2% de crocines, en poids par rapport au poids total de l’extrait deCrocus sativuset/ou stigmate deCrocus sativus, mesuré par méthode HPLC.

- Composition selon l’une des revendications précédentes, caractérisée en ce que l’agent antioxydant représente entre 100 ppm et 5000 ppm en poids par rapport au poids total de stigmates deCrocus sativus .
- Composition selon l’une des revendications 3 à 11, caractérisée en ce que l’agent chélateur est choisi parmi l’acide citrique ou l’EDTA et des protéines
- Composition selon l’une des revendications précédentes, caractérisée en ce que le stigmate deCrocus sativuset/ou l’extrait deCrocus sativusest encapsulé ou microencapsulé.
- Utilisation d’un agent antioxydant pour lutter contre la dégradation des molécules présentes dans le safran et/ou réduire l’instabilité des molécules présentes dans le safran, caractérisé en ce que ledit agent antioxydant représente entre 0,01% et 25% en poids par rapport au poids total de la matière sèche de crocines présentes dans le safran, mesuré par HPLC.
- Utilisation selon la revendications précédente, caractérisé en ce que l’agent antioxydant permet de limiter la dégradation des crocines et/ou du safranal.
