Les sérums ont été prélevés régulièrement et la détection d'anticorps anti-VP7 et anti-NS1 a été effectuée par méthode ELISA de compétition. La détection d'anticorps anti-VP7 a été réalisée à l'aide du kit commercial BTC d'IDVET. La détection d'anticorps anti-NS1 a été menée par méthode ELISA direct de compétition à l'aide des anticorps 11G1 ou 9B4 couplé à la peroxydase et selon le protocole décrit ci-après. Cinquante microlitres de sérum dilué au en solution SL ont été déposés par puits sur une plaque présensibilisée à l'aide de protéine NS1 native. La séroconversion anti-NS1 n'apparaît qu'après la 2è' injection vaccinale avec une diminution à 40 % et 44 % du signal de l'anticorps monoclonal 11G1 et 9B4, respectivement, entre J+38 et J+73. Un différentiel s'établit entre les deux types d'anticorps monoclonaux anti-NS1 après la 3e injection vaccinale. En effet, le signal diminue de 51 % à J+74 à 16 % à J+108 pour l'anticorps 1 1G1. Dans ces conditions, les limites du potentiel DIVA de l'anticorps monoclonal 11G1 semblent donc atteintes. Au contraire, le signal de l'anticorps 9B4 reste stable aux alentours de 48 %. Aux vues de ces résultats, il apparaît que l'anticorps monoclonal 9B4 est plus performant en DIVA BTV que l'anticorps monoclonal 11G1. Cinquante microlitres d'anticorps monoclonaux anti-NS1 (8 µg/mL en PBS) ont été déposées dans des plaques 96 puits puis incubées à température ambiante sur la nuit.
