The present invention relates to the synthesis of novel complexes of activated and recyclable ruthenium 5 I for olefin metathesis: Ls R./Ne, R 10 in which R s Alkyl, aryl • X Halogene, OR, OCOR • RI = Alkyl, perfluoroalkyl, aryl • R2 Alkyl, perfluoroalkyl, aryl ... RI and R2 can form a ring between them. It relates more particularly to the synthesis of ligand of styrenylether type having an amide function and their complexations with ruthenium derivatives (Grubbs II catalyst or UMICORE catalyst). When the acetamide function has a perfluorinated methyl (trifluoromethyl), a strong activation of the catalyst is observed, characterized by relatively high conversions in very short times. Under these conditions, an economic repercussion can be envisaged by means of a significant reduction in the catalytic load in the metathesis reactions (without altering the yields). This activating acetamide function can also serve as a spacer for the introduction of an ionic unit. The final objective being to allow better recycling of the catalytic complex (expensive) in aqueous / ionic solvents or on a solid support (continuous flow reaction) and to obtain a clear reduction in the cost of the reaction while at the same time avoiding contamination of high added value products (pharmaceutical molecules). Recyclable or activated ruthenium-based complexes for the olefin metathesis reaction have been the subject of several patents as well as some publications in scientific journals. The development of these complexes is based on the previous work of R. Grubbs (Caltech, University of California, USA) relating to the ruthenium 2a-b complex (pre-catalyst). 2b Thus the appearance of the first recyclable complex 3a-b with a styrenylether ligand (so-called bommerang ligand) was described in 1997 (work by Hoveyda, Boston University, USA). A worldwide patent was published in August 2000 (WO0214376). The direct advantage of this complex is that it allows recycling of the precatalyst which is recovered at the end of the reaction and can be reused. However, significant losses (up to 10% per cycle) are observed. The second advantage is to minimize the presence of metallic waste (toxic) in the reaction products. However, this complex turns out to be less active than that of Grubbs II 2b.
The first activated complex 4 was described in 2002 based on the electronic effect induced by the presence of a nitro group (NO2) on the styrenylether ligand of Hoveyda 5 (previously described). The activation of this precatalyst is based on the strongly accelerated detachment of the styrenylether ligand which causes a rapid initiation of the catalytic cycle and therefore a significant reduction in the reaction time. The reactions can then take place under milder conditions (room temperature) with lower catalytic loads. This activated complex was patented in 2002 by Boehringer Ingelheim 10 international (WO2004035596). However, this complex is not easily recycled, which furthermore leads to significant contamination of metallic residue (ruthenium) in the reaction products. NO2 W02004036596 1-1 Mes.NY N.Mes CL 'Cl / Ru_ 4 15 A clear improvement relating to the use of recyclable complexes for olefin metathesis lies in the control of good reactivity without altering the recycling of the catalyst . Indeed, reactivity and recycling are two opposing properties characterizing this type of complex. The activity of the catalyst (catalyst 3) can easily be increased, but this is done to the detriment of recycling. Conversely, the catalyst can be recycled suitably, but this comes to the detriment of the reactivity of the catalytic species (catalyst 2 less active than 1). It is therefore necessary to optimize this balance between activity and recycling and to determine the best compromise between these two opposing properties.
The aim of the present invention is to promote ruthenium complexes by optimizing the compromise between these opposing properties: these new catalysts will be capable of combining excellent activity while retaining good recycling. Under these conditions, a reduction in the catalytic load for optimal activity and recycling could be envisaged, irreparably leading to a large reduction in metal waste in the final products. The introduction, according to the invention of an amide function on the styrenylether ligand appears to be an element of choice for obtaining a good compromise between activity and recycling because it has the dual characteristic of (i) promoting the activation of the catalyst and of (ii) serve as a spacer for the introduction of an ionic tag with a view to immobilization in the aqueous and / or ionic phase as well as on a solid support. The final objective of this invention lies in obtaining products with high added value with very low ruthenium contents (<10-20 ppm). Synthesis of the complexes The complexes la-d possessing an amide function are obtained in two steps from the functionalized aniline 5 (synthesized in 4 steps from the para-nitro-phenol, according to the process described in "Activated pyridinium-tagged ruthenium complex as efficient catalyst for Ring-Closing Metathesis. The results (see FIG. 1) clearly show the activating effect of the acetamide function, in particular when the latter has a trifluoromethyl group (complex lb) with more than 37% conversion after only 15 min of reaction against 5% in the case of of the Hoveyda 3b complex.
The inventor also studied the influence of the activation of trifluoroacetamide on the complex carrying it an IMes ligand ( resulting from complexation with the Nolan catalyst) instead of a SIMes ligand. To their surprise and for the first time, a similar activity is observed (see figure 2) while a clear difference exists in the non-activated version [the Grubbs II 2b catalyst (SIMes ligand) is much more active than 10 the Nolan catalyst (IMes ligand)]. This result is very encouraging because the catalytic species carrying an IMes ligand (obtained from the precatalyst of Nolan) is much more thermally stable than the catalytic species carrying a ligand SIMes (generated from the precatalyst of Grubbs II 2b). It would then be possible to carry out olefin metathesis reactions under more stringent conditions (high and prolonged heating) with the activated complex when the substrates are highly hindered (eg: tetrasubstituted olefins).
Sa complexation avec le catalyseur de Grubbs II conduit au complexe 11.The trifluroacetamide function being clearly identified as being the function most capable of activating the precatalyst, the introduction of an ionic unit (ionic tag) can then be carried out. For this, the present invention proposes to introduce via the amide a chloroperfluoroalkyl group in order then to allow substitution of the chlorine atom by a tertiary amine (imidazole, pyridine, etc.). Thus, the inventors have performed the substitution with dimethylimidazole on chorodifluoroacetamide 9 to easily lead to the desired ionic ligand 10. Its complexation with the Grubbs II catalyst leads to complex 11.
