Abstract
Description
TYPE 1 R-PHYCOERYTHRIN, A PROCESS FOR ITS
OBTAINING AND USES THEREOF
The present invention relates to a new type of orange-red dye, a process for obtaining it, and its uses.
For a few years now, the use of dyes has been highly regulated, whether in the field of cosmetics, dietetic products or pharmaceutical compositions. In fact, the requirements, in particular as regards the non-toxicity of these dyes, have been severely increased.
This is why there is a growing tendency to use natural dyes.
This is how we have already proposed to extract a dye from certain red algae. In particular, dyes from the phycoerythrin family, photosynthetic pigments of protein nature, have already been extracted from these algae. However, these dyes have a serious drawback due to their lack of stability. Indeed, they quickly lose their bright and strongly fluorescent color. This is why we are obliged, most often, to keep them away from light.
The applicant has found that it is possible, against all expectations, to extract a dye from the R-phycoerythrin family of type 1 which is stable, that is to say which does not lose its color even after a long period storage in daylight, from a particular variety of red algae.
This orange-red dye, consisting of an R-phycoerythrin type 1 is characterized by the fact that it is extracted from the alga Dilsea carnosa.
- maximum d'excitation du spectre d'émission en fluorescence = 498 - 502 nm.Advantageously, this dye has the following parameters
- maximum absorption in the ultraviolet and visible spectrum = 498 - 500, 538 - 540 and 565 - 567 nm
- maximum emission of the fluorescence excitation spectrum = 575 - 577 nm
- maximum excitation of the fluorescence emission spectrum = 498 - 502 nm.
This process includes the steps of:
a) macerate thalli of Dilsea carnosa
b) filter the macerate from step a)
c) centrifuge the filtrate for 20 to 40 minutes
d) recover the supernatant from the centrifugation and introduce ammonium sulfate therein
e) centrifuge the solution for 20 to 50 minutes
f) take up the centrifugation pellet with water and dialyze it for 24 to 48 h.
According to other advantageous characteristics of the process
- at stamp a), the thalli are macerated in about twice their weight of water
- in step a), maceration is continued for 4 to 8 weeks
- in step d), add ammonium sulfate at 30-60% saturation
- At the end of step g), the dye is recovered in aqueous solution or a lyophilization of the latter is carried out in order to obtain a dry extract.
Finally, the invention relates to the use of this dye for the preparation of a cosmetic, dietetic, food or pharmaceutical composition and as a tracing agent in immunofluorescence.
Other characteristics and advantages of the invention will appear from the description of an embodiment of the invention, made with reference to the accompanying drawings in which
- Figure 1 is a representation of the absorption spectrum in the ultraviolet and the visible of the dye of the invention
- Figure 2 is a representation of the emission and excitation spectra in fluorescence, of this same dye.
The red alga Dilsea carnosa is an alga from the branch of the Rhodophytas which occurs from Greenland to the Vendée coasts and then reappears on the Cantabrian coast after interruption on the Basque coast.
It has a dark red thallus in the form of thick, fleshy blades that can reach 50 to 70 cm in length and 20 to 30 cm in width.
As mentioned above, dyes have already been extracted from red algae. However, these dyes are not stable and there is a prejudice in scientific circles against these dyes.
Contrary to all expectations, it turned out that the orange-red dye extracted from Dilsea carnosa is perfectly stable, even in daylight.
We will describe below a preferred procedure for the extraction of the dye from the alga Dilsea carnosa.
The harvested algae are washed carefully in order to rid them of their epiphytes which may interfere with the quality of the dye.
They are then preferably rinsed thoroughly with distilled water.
A determined quantity of algae is macerated in twice its weight of water, the fronds having to bathe in the liquid. Advantageously, distilled water is used. The container can then be placed in a medium at low temperature, for example a refrigerator, because of the too strong microbial proliferation at room temperature, for a period of 4 to 8 weeks.
After this period, the dye still continues to diffuse but its recovery is much more difficult, still due to microbial development.
The maceration juice is then preferably filtered, for example on cotton to remove the largest cellular debris, then the filtrate is centrifuged at approximately 3000 rpm for 20 to 40 minutes, this step making it possible to remove the smaller cellular debris. .
The supernatant is then recovered and then added to the solution of ammonium sulphate (30 to 60% saturation) which has the purpose of precipitating the proteins constituting the R-phycoerythrin type 1 which constitutes the dye.
The solution is then centrifuged for 30 to 50 minutes. Then the supernatant, which contains the majority of germs and which has a very strong odor, is eliminated. The pellet is then rinsed lightly with distilled water then resolubilized always in distilled water.
The solution is dialyzed for 24 to 48 hours in order to remove all the salts present and in particular ammonium sulphate.
To do this, a dialysis rod trampering in distilled water is used. Preferably, a sterilizing filtration of the dialysate is carried out.
The extract can then be used as it is, in aqueous solution.
It can also be subjected to a dehydration operation, for example by lyophilization, to obtain a dry product.
The yield of the extraction operation is at least of the order of 1% of purified dry extract relative to the dry weight of algae.
The dye obtained has a very intense orange-red color with a clear orange fluorescence.
The product obtained was subjected to several tests in order to be able to define some of its properties.
Thus, solubility, stability and absorption tests were performed.
I - TESTS DE SOLUBILITE
- solubilité - dans l'eau = totalement soluble
- dans l'éthanol (95 ) = insoluble ; formation d'un
précipité
- dans la glycérine et le propylène glycol =
insoluble
- dans les principales huiles végétales et
minérales = insoluble.I - SOLUBILITY TESTS
- solubility - in water = fully soluble
- in ethanol (95) = insoluble; formation of a
precipitate
- in glycerin and propylene glycol =
insoluble
- in the main vegetable oils and
mineral = insoluble.
- variations in pH: stability of pH 4.5-5 at pH 10
- temperature variations: stability up to 60 C
These properties make it possible to use the dye in numerous compositions, in particular cosmetic and pharmaceutical.
III - TESTS D'ABSORPTION
Analyzes were carried out in the ultraviolet and visible light domain. Emission and fluorescence excitation analyzes were also carried out on the dye.
A UV-Visible spectiophotometer of the brand HITACHI, model U2000 was used at a temperature of 20 C. For fluorescence, a spectrofluorimeter SCM 4800 was used, at a temperature of 20 C.
Figures 1 and 2 attached are representations of the corresponding spectra.
Thus, curve 1 corresponds to the measurement of the absorption of the dye in the range of ultraviolet and visible light. The absorption maxima are in the ranges 498 - 500, 538 - 540 and 565 - 567 nm.
Curves 2 and 3 correspond to the fluorescence excitation spectra of the dye, with an emission at 575-577 nm.
Curve 4 represents the fluorescence emission spectrum, with an excitation at 498 - 502 nm.
Because of its qualities of stability and solubility, the product of the invention has many possible applications as a dye.
In addition to the uses already mentioned in the cosmetic and pharmaceutical industries, it can also be used as a food coloring, including for dietetic products.
In addition, because of its strong fluorescence, it could be used as a tracing agent in immunofluorescence.
