The invention relates to bioavailable α-lino acid complexes (enic, plant extracts containing them and pharmaceutical compositions incorporating them.
X-linolenic acid is very unstable so that it is not bioavailable when administered to a living being, particularly a mammal including a human.
It is an acid essential to the metabolism of the human being which is not generated by it and which must be brought by an external source. The main problem is to provide cclinolenic acid in a bioavailable form.
The invention aims to solve a new technical problem consisting in providing a formulation of bioavailable α-linolenic acid having increased bioavailability which is simple to prepare and which is sufficiently stable over time to allow therapeutic use. in the best conditions.
The invention solves this problem in an extremely simple way by providing a complex form of oc-linolenic acid which unexpectedly exhibits remarkable bioavailability.
Thus according to a first aspect, the invention provides a complex of orlinolenic acid characterized in that crlinolenic acid is complexed or combined with a phospholipid.

According to a particularly advantageous embodiment of the invention, the alpha-linolenic acid complex combined with the phospholipid is combined with an alkaline or alkaline-earth ion, in particular sodium, potassium, calcium or magnesium. According to the invention, calcium and magnesium are preferred.
This complex could be prepared synthetically from alpha-linolenic acid and the corresponding phospholipid which would be reacted under operating conditions preserving the chemical structure of α-linolenic acid.
However, this complex is found naturally in plants and can be obtained from sources constituted by plants which are subjected to specific extraction treatments which will be described later.
The particularly preferred plants used as raw material for the isolation of the complex according to the invention are those of families

Ribes, etc.
The present invention relates, according to a second aspect, to a method of isolating the x-lin6lenic acid phospholipid complex according to the invention from an appropriate plant, characterized in that it comprises
- the extraction of part or all of the plant with a polar or non-polar solvent
- subjecting the crude extract obtained to one or more appropriate separation steps. Advantageously, one of these separation steps comprises subjecting the crude extract to chromatography on a column of silica or alumina
Any polar or non-polar solvent well known to those skilled in the art can be used. As a polar solvent, water alone is not suitable, but a mixture of water with an alcohol miscible at the rate of one part of alcohol per 2 to 20 parts of water constitutes a good solvent. extraction using heat should be avoided. In general we therefore work at room temperature. The particle size as well as the contact times are determining parameters and the person skilled in the art knows after a few simple tests to determine the particle size and the contact time which are satisfactory for the extraction from a given plant. Counter-current extraction techniques are preferable as much as they allow milder isolation conditions. Likewise, lyophilization of the extract is preferred to any other form of separation of the extract. From the examples of extraction which are given below, the person skilled in the art is able to implement the isolation process according to the invention.
The extraction procedures described in the examples form an integral part of the present invention.
According to a third aspect, the invention also relates to a pharmaceutical composition, characterized in that it comprises, as active principle, a complex of α-linolenic acid and a phospholipid, preferably as previously defined, incorporated in a pharmaceutically acceptable carrier or vehicle excipient.
According to an alternative embodiment, the concentration of complex according to the invention is between 1 mg and 50 mg by weight per unit dose.
According to a particular embodiment of the invention, the pharmaceutical composition is characterized in that it is a composition with anti-inflammatory activity. This composition can be formulated in the usual way; in particular, a formulation can be produced for oral, topical administration for local or percutaneous use.
According to another particular embodiment of the invention, the pharmaceutical composition is characterized in that it is a pharmaceutical composition with analgesic activity, in particular formulated for oral, topical administration for local or percutaneous use. .
According to a third particular embodiment of
The invention, the pharmaceutical composition is characterized in that it is a pharmaceutical composition with antipyretic activity, in particular formulated for oral, topical administration for local or percutaneous use.
It has also been observed completely unexpectedly that this complex according to the invention has an action on the mechanism of prostaglandins and of the entire cascade of arachidonic acid, in particular on the cycles of inositolphosphate and phosphatidylinositol -phosphate, which helps restore basic metabolism.
The invention therefore makes it possible to treat all the disturbances resulting from a dysfunction of the metabolism of prostaglandins or of arachidonic acid.
The present invention also covers a process for the preparation of a pharmaceutical composition, characterized in that a complex of x-linolenic acid and a phospholipid, in particular as defined above, is incorporated in a pharmaceutically carrier or excipient vehicle acceptable.
Finally, the present invention also covers a method of therapeutic treatment of a mammal including a human being, characterized in that this mammal is administered a therapeutically effective amount of an α-linolenic acid complex and a phospholipid, in particular as previously defined.
According to particular variants of this therapeutic treatment method, an anti-inflammatory treatment or an analgesic treatment or an antipyretic treatment is carried out.
The present invention will now be illustrated from several examples of the invention given simply by way of illustration and which therefore cannot in any way limit the scope of the invention. In the examples, as in the text and in the claims, all the percentages are given by weight, unless otherwise indicated.
