Abstract
Description
The present invention is in the field of medical analyzes and it relates to a blood collection tube intended for analyzes of blood serum, as well as a particular treatment of the tube or of its constituent material entering into its manufacturing process.
We recall that we have long used small glass tubes to store the blood collected and make it clot; then because of the increasing number of analyzes, at the cost of glass tubes and their fragility, we replaced the glass with plastic (polyethylene, polypropylene, polystyrene, etc.) we then noticed that the blood coagulated a lot slower and with less good clot retraction than in glass tubes; it was then assumed that the oxy-silicic compounds of glass could be responsible for good coagulation, for example could catalyze fibrino-formation. We then thought of introducing into the plastic tube after its manufacture for example, the glass powder, and good coagulation was found; however, the glass powder obstructed or deteriorated the complex serum analysis apparatus; therefore, the introduction of glass powder had to be practically abandoned. It has been proposed to introduce organic accelerators of coagulation into the tube, but we have never obtained, while complicating the analysis process, as good results as with the old glass tubes.
Finally, it should be recalled that the collection tubes intended for the analysis of serum incorporate a certain number of plastic granules, the density of which is greater than that of the serum but less than that of the coagulum (clot) and which are intended to both improve coagulation and promote separation of serum and clot.
Starting from the idea that certain silicon derivatives were responsible for the coaguant effect, we tried to treat the tubes or the granules with the silicone and silane compounds; the result was negative.
The object of the present invention is to propose a treatment for the plastic tube and / or its constituent material, giving them coagulating properties as good as those of glass tubes, taking up the idea that the oxidative compounds silicics must have been present in the tube.
According to the present invention, a collection tube, intended in particular for the analysis of blood serums, said tube being of the kind made of plastic material, which can contain plastic granules and include a stopper, is generally characterized by the presence, at least in trace amounts, in the material of the tube or of the granules which it may contain, or on their surface, of an oxidized derivative of silicon (-Si-O-), excluding silicones and silanes; preferably said derivative is a silicate derivative, for example a kaolin which, advantageously, will have been washed in an acid medium.
The invention also relates to manufacturing processes, or treatment entering into the manufacturing process of the above-defined tubes. The process generally consists in placing the oxidized derivative of silicon in solution or in suspension in a surfactant liquid, advantageously a surfactant compound, for example a polyhydroxylated compound; then placing this composition (solution, suspension) in the presence of either the tube itself and / or its granules, or the raw material, generally presented in the form of granules, intended for molding or extruding the tube. Thanks to the surfactant, the oxidized derivative covers the surface of the plastic; a monomolecular layer is sufficient since it only acts as a catalyst.
Experience has shown that in the case where the granules are coated with raw material, the coagulating effect which was thus conferred on them is found in the molded tube; it has even been found that it is sufficient that a small fraction of the granules has been coated with the composition, for the coagulating effect to be satisfactory.
A suitable surfactant is polyethylene glycol, but any other surfactant, anionic, cationic or nonionic could be suitable provided that it does not have an inhibitory action by itself on coagulation, or disruptive action on subsequent serum analyzes.
A method which has given good results comprises the following stages - firstly mix 10 g of acid-washed and suitably dried kaolin with 100 ml of polyethylene glycol - secondly sprinkle with the preceding suspension 10 kg of granules of the material -plastic intended for the molding of the tubes, and mix according to the technique known as "rotating barrel" the suspension and the granules, so that the suspension covers without excess the surface of the granules - remove 1 kg of these granules to constitute the granules present in the tube, mix the 9 kg of granules remaining with 100 kg of granules intended for molding the tubes.
It will be possible, if necessary, to recognize that a sampling tube corresponds to the invention by subjecting a certain quantity of tubes and granules ground to a thorough leaching and by subjecting the concentrated leachate then filtered to a chemical analysis in order to reveal the presence of the group (-Si-O-).
Although a particular embodiment of the invention has been described, both in the product concerned and in the process for obtaining it, it should be understood that the scope thereof is not limited to the examples cited but that it extends to any sampling tube comprising the general characteristic stated above and to any process falling within the general definition which has been given thereof.
