Abstract
Description
The present invention relates to new pharmaceutically acceptable microemulsions in particular for parenteral administration.
Therapeutically active products have physical or biological properties which can either prevent their administration by certain routes, or cause harmful side effects which must be eliminated or reduced. However, certain products, because of their low solubility in physiologically acceptable solutions, cannot be administered parenterally; others have harmful effects on the mucous membranes (stomach, esophageal, intestinal, rectal, ...) with which they are in contact. More particularly, the existence of an injectable form of a therapeutically active substance is of considerable interest in terms of bioavailability or in terms of the adaptation of the route of administration to the treatment of the disease considered. of an injectable composition, for example intravenously, is linked to the solubility of the pharmacologically active substance in an essentially aqueous medium. For example, active substances with a low solubility in water and at a concentration below 0.1 mg / cm3 cannot be used for the preparation of an injectable form when the active unit dose is l 'order of 100 ring. It is therefore important, in order to prepare injectable formulations containing a sufficient quantity of active product, to be able to increase the solubility of the active substance in the pharmaceutically adaptable medium.
One of the methods used for parenterally administering pharmacologically active substances having insufficient solubility in aqueous media consists in dissolving the active substance in an appropriate oil, then in producing an emulsion with an aqueous continuous phase which constitutes an opaque oil-in-water system. water formed from water and organic substance in the presence of one or more emulsifiers.
Another commonly used method is to administer the active substances in the form of an injectable suspension.
However, these formulations have drawbacks such as the thermodynamic instability of the emulsions or the obtaining of particles of irregular size.
Microemulsions can constitute systems making it possible to increase the dissolved amounts of pharmacologically active substances which are poorly soluble in aqueous media.
A microemulsion is a dispersion of two immiscible liquids in which the particles of the dispersed phase have an average diameter of less than about 1400 A. This colloidal dispersion of particular structure is obtained using one or more emulsifiers which stabilize the system .
Beyond 1500 A begins the field of conventional thermodynamically unstable white-milky emulsions, while,
o below 100 A, lies the domain of transparent and thermodynamically stable micellar solutions. Also the field of microemulsions is at the border of conventional emulsions and micellar solubilization and, due to the very small size of the particles of the dispersed phase, these systems are transparent.
Qualitatively, a microemulsion comprises a hydrophilic phase, a lipophilic phase and one or more emulsifiers ensuring the formation of an interfacial film which separates the continuous and discontinuous phases.
The choice of the emulsifying mixture, the constituents of which must be carefully balanced, conditions the formation of the microemulsion structure, characterized mainly by its capacity to simultaneously dissolve large quantities of water and oil to form a homogeneous, transparent, isotropic and thermodynamically stable liquid. .
The micromulsions can be classified into two groups comprising the microemulsions with aqueous continuous phase called "oil in water" and the nicroemulsions with continuous oil phase called "water in oil".
The mi cro enulsions most commonly used in the industrial field consist of an oil, water and two surfactants which are
a) a surfactant (also called surfactant which is an amphiphilic hydrophilic tendency which can be ionic (sodium lauryl sulphate, potassium oleate) or non-ionic (polyoxyethylenated sorbitan esters)
b) a cosurfactant (also called cosurfactive) which is an amphiphile with a lipophilic tendency which is generally chosen from alcohols containing 4 to 14 carbon atoms and nonionic agents such as sorbitan monolaurdte or tetraoxyethylene glycol dodecyl ether.
There also exist microemulsions containing a single surfactant which is generally chosen from fluorinated ionic surfactants or polyoxyethylenated fatty alcohols.
Microemulsions have been proposed for a pharmaceutical or cosmetic application.
Thus, in particular according to European patent 51,526, micro-emulsions of fluorocarbons have been described as blood or plasma substitutes because of their ability to transport oxygen, nitrogen and carbon dioxide or as a medium for preserving isolated organs, or even as an infusion vehicle.
According to American patent US 4 146 49cl, microemulsions have been proposed as a vehicle for lipophilic active principles which can be administered orally or by injection or as an intermediate medium allowing stable microdispersions of hydrophobic active principles to be obtained after evaporation of the oily phase. The microemulsions are then used as transient phases making it possible to increase the quantity of lipophilic active principle solubilized in the micelles of emulsifiers.
In French patent FR 2 502 951 are described liquid or semi-solid microemulsions intended for the systemic or local administration of active principles by cutaneous life.
Finally, numerous microemulsions have been described as vehicles for cosmetic preparations.
However, these various microemulsions are not particularly suitable for parenteral administration of therapeutically active substances because of their high content of surfactants or of their narrow zone of thermal stability.
I1 has now been found, and this is what is the subject of the present invention, that pharmaceutical injectable compositions can be prepared in the form of microemulsions having emulsifier contents compatible with parenteral administration and allowing the incorporation of therapeutic doses of poorly water-soluble active ingredients.
The pharmaceutically acceptable microemulsions according to the present invention are composed of water optionally containing an electrolyte, an oil, a surfactant and a cosurfactant and optionally one or more organic or inorganic substances pharmacologically active or not.
The constituents must be pharmaceutically acceptable, that is to say non-toxic for the organism at the concentrations employed.
The choice of the constituents making it possible to produce a microemulsion is guided by the consideration of various parameters among which the most important are the physicochemical nature of the oil to be emulsified (polarity, viscosity, solvent property with respect to the active substance to be incorporated), the composition of the emulsifying mixture (cosurfactive, surfactive), the chemical composition of the aqueous phase and the temperature.
The oil is generally made up of an ester of an aliphatic alcohol containing 2 to 22 carbon atoms or of an aromatic alcohol with an aliphatic carboxylic acid containing 10 to 22 carbon atoms or with an aromatic acid such as the oleate ethyl, ethyl laurate, hexyl laurate, isopropyl myristate or benzyl benzoate.
The surfactant is of the ionic type and it can be chosen from the salts of aliphatic acids containing 10 to 22 carbon atoms with organic or mineral bases such as sodium or post potassium oleate, triethanolamine or diethanolamine oleate .
The cosurfactant is an aliphatic polyol containing 3 to 18 carbon atoms such as 2-ethylhexanediol-1,3 or an aromatic alcohol such as benzyl alcohol.
The electrolyte optionally contained in the aqueous phase is chosen from pharmaceutically acceptable mineral salts such as sodium chloride or disodium phosphate.
The organic or inorganic substances which are pharmacologically active or not are, for example, ethanol, urea, lactose, sucrose or therapeutically active molecules which are sparingly soluble in water.
Microemulsions for pharmaceutical use must constitute translucent, isotropic and thermodynamically stable systems having high water and oil contents and be physiologically acceptable for the organism. Furthermore, the microemulsions must be able to be diluted in physiological aqueous media such as plasma, which implies the production of systems with an aqueous continuous phase.
The microemulsions according to the present invention are obtained by stirring the mixture of the various constituents in proportions previously determined by routine tests, at a temperature in the region of 200C.
To obtain pharmaceutically acceptable microinulsions, it is advisable to produce a composition in which the aqueous continuous phase contains a maximum of dispersed oily phase for a minimum of surface-active agents.
The choice of the constituents and the determination of their relative proportions are carried out by the use of ternary diagrams established on the basis of a titration method. Generally speaking, the determination methodology. optimal conditions has three successive phases which include
1) the selection of the emulsifying systems (surfactant and cosurfactive) making it possible to microemulsify a specific oil which is chosen according to the solubility criteria of the active ingredient to be incorporated and the approximate determination, for favorable systems, of the cosurfactive / surfactant and surfactant / water necessary to obtain microemulsions,
2) optimization of the microemulsions obtained during the first phase carried out by seeking the values of the cosurfactive / surfactive, surfactant / water and electrolyte / water ratios which characterize the optimal microemulsion, that is to say the microemulsion having a content maximum in oil for a minimum quantity of emulsifiers.
3) the study of the ternary diagram grouping together all the combinations achievable with the following three basic constituents - oil - the aqueous phase which consists of water with electrolyte added at the optimal concentration determined during the second phase - the emulsifying mixture (surfactant and cosurfactive) with an optimal cosurfactive / surfactant ratio determined during the second sentence.
This type of diagram makes it possible to view, for a constant value of the cosurfactive / surfactant and electrolyte / water ratios, the extent of the zone of microemulsions forming for increasing contents of emulsifying mixture in the system, that is to say for increasing surfactant / water ratios.
The ternary diagram thus produced makes it possible to follow the maximum quantity of incorporable oil as a function of the content of system emulsifiers.
Carrying out these three successive steps leads to an optimized formula, that is to say to a microemulsion with an aqueous continuous phase containing a maximum of dispersed oily phase for a minimum of surfactants.
Application of the method described above to a system consisting, for example, of ethyl oleate (oil), distilled water containing an electrolyte, triethanolamine oleate (surfactant) and alcohol benzylic (cosurfactive), shows that - the cosurfactive / surfactive ratio must be between 0.7 and 1 - the quantity of oil that can be incorporated is a linear function of the rap
surfactant / water port - the addition of electrolyte to the aqueous continuous phase allows
significant increase in the solubilization capacity of the system,
the quantity of electrolyte necessary to obtain the optimum
varying from one salt to another and being a function of the ionic strength
salt considered. For example, in the above system, the con
sodium chloride concentration is close to 2% and that of
disodium phosphate is close to -6 by weight of the aqueous phase.
For the system considered, the ternary diagram making it possible to produce the optimal microemulsion is represented by FIG. 1.
Examination of the ternary diagram shows that - the extent of the domain of monophasic systems is very important
(upper part of the diagram) - for the salt concentration used, there is a growth
from the field of microemulsions. The exact location of this pro
along the monophasic domain present for the realization-of
the microemulsion is of considerable importance because the more this excess
sance is important the higher the emulsifier content of microemul
corresponding is weak.
Thus for this system, the terminal portion of the substance (portion closest to the axis indicating the proportions in aqueous phase) corresponds to microemulsions having relatively low emulsifier contents, of the order of 8%. for an oil content of around 4 m.
In this way, it is possible to prepare an opalescent and stable mixture, the composition of which is as follows
- ethyl oleate 4%
- triethanolamine oleate 4%
- benzyl alcohol 4!
- distilled water 86.42%
- sodium chloride 1.58%
At this stage of the formulation, a final optimization of the cosurfactant / surfactant ratio can be carried out in order to reduce the proportion of cosurfactant contained in the composition described above.
To this end, it is necessary to seek the smallest value of the cosurfactive / surfactant ratio allowing the obtaining of a transparent and stable microemulsion containing 4% of ethyl oleate for a triethanolamine oleate content equal to 4%.
In this way, it is possible to prepare an opalescent and stable mixture, the composition of which is as follows:
- ethyl oleate 4%
- triethanolanine oleate 4%
- benzyl alcohol 3%
- distilled water 87.4%
- sodium chloride 1.6%
The ability of this system to form microemulsions with an aqueous continuous phase is essential because it allows the use of sufficient amounts of surfactant without reaching toxic concentrations of cosurfactant.
The microemulsion thus obtained has a limpid, opalescent appearance. Its pH is close to 8.1.
The particle size, determined by quasielastic light scattering, is close to 200 A and the polydispersity factor is close to 0.1.
This microemulsion is stable at different temperatures and its fundamental properties are not altered after 3 months at 4 C, 250C, 350C and 45du.
Furthermore, this microemulsion can be diluted in an infusion solution. It remains stable for 24 hours after 1/10 dilution and for 72 hours after 1/100 dilution, which allows, in particular, administration after dilution in physiological saline.
Other pharmaceutically usable microenJlsions can be obtained by replacing the ethyl oleate with other esters such as isopropyl myristate or benzyl benzoate.
However, each system constituting a particular case it is necessary to apply the methodology previously described to obtain an optimal formulation.
Microemulsions with an aqueous continuous phase may be useful as pharmaceutical vehicles for lipophilic or amphiphilic active principles such as hormones, fat-soluble vitamins or any other non-water-soluble active substance.
