Abstract

Adsorption material, esp. for adsorption of phosphates contained in the blood, comprise porous beads containing zirconium oxide and coated witha permeable, hydrophilic polymer of low mol. wt. cpds. Before coating, the beads pref. comprise 20-100 wt.% zirconium specific surface area of 500-1300 sqm/g. The pref. coating is collodion, of about 0.05 micron thickness. Adsorbent cartridges containing the above material are also claimed. Treatment of patients suffering from chronic renal insufficiency, so as to reduce blood phosphate levels during extracorporal blood treatment. The treatment may be effected on the whole blood, the plasma or on a haemofiltrate prior to reinjection.

Description

The present invention relates to an adsorbent material intended in particular for the adsorption of phosphates contained in the blood.

- l'hémodialyse, qui consiste à faire passer les substances à éliminer à travers une membrane semiperméable;
- l'hémoperfusion, qui consiste à faire adsorber certaines substances sur un matériau adsorbant;
- l'hémofiltration;
- l'hémodiafiltration;
- la dialyse péritonéale;
- la séparation plasmatique.
Numerous methods of extra-corporeal blood treatment are known which are used for the treatment of patients suffering from chronic renal failure. These methods include
- hemodialysis, which consists in passing the substances to be eliminated through a semipermeable membrane;
- hemoperfusion, which consists in adsorbing certain substances on an adsorbent material;
- hemofiltration;
- hemodiafiltration;
- peritoneal dialysis;
- plasma separation.

In most of these methods, the phosphate content is not reduced enough. However, it is known that high levels of phosphates in the blood cause metabolic disturbances.

In addition, conventional adsorbents (such as activated carbon) do not allow a sufficient lowering of the phosphate levels.

The present invention aims to provide an adsorbent material making it possible to sufficiently lower the levels of phosphates contained in the blood.

To this end, the present invention relates to an adsorbent material consisting of porous beads containing zirconium oxide and coated with a thin film of a hydrophilic polymer permeable to low molecular weight compounds.

These porous beads advantageously contain from 20 to 100% by weight of zirconium oxide.

These beads advantageously have an average particle size of 100 to 900 microns, preferably 300 to 600 microns, and a specific surface before coating of 500 to 1300 m2 / g.

The porous beads used in the present invention can be manufactured by mixing powdered zirconium oxide (advantageously having a particle size of less than 20) with an organic binder which can be decomposed thermally, formation of beads or particles from the mixture obtained and heating at a temperature sufficient to decompose the binder and provide beads composed of zirconium oxide and carbon or even of zirconium oxide alone (by decomposition in an oxidizing atmosphere).

The organic binder may contain, in addition to carbon and hydrogen, oxygen and nitrogen, but must be free of contaminants such as sulfur and chlorine.

Un procédé de préparation de perles poreuses qui peut Stre utilisé à out effet est décrit dans le brevet US 4 256 676. A process for preparing porous beads which can be used for any purpose is described in US Pat. No. 4,256,676.

According to this process, porous beads containing a metal oxide are obtained by mixing the powdered metal oxide with an organic binder which can be decomposed thermally. This binder can in particular be a thermopolymerizable monomer, such as styrene, divinyl benzene, furfuryl alcohol or a liquid phenol formaldehyde resin.

Porosity adjusting agents can also be incorporated into the mixture, for example cellulose, wood flour, starch or sugar.

The mixture is then formed into beads by passing through a sieve having an appropriate mesh size. Then, the beads are heated to a temperature sufficient to polymerize or crosslink the binder.

The beads are then heated to a temperature of up to 3500C to decompose the binder and obtain a composite material consisting essentially of carbon and metal oxide.

Heating to 9000C in an oxidizing atmosphere makes it possible to obtain pearls consisting solely of metal oxide.

The beads based on zirconium oxide which can be obtained in this way are then coated with a thin film of a hydrophilic polymer permeable to low molecular weight compounds (that is to say in particular permeable to phosphate ions).

This coating can be, in particular, a thin film obtained by application of a collodion, in particular by the technique of microencapsuîation by evaporation. The collodion used can for example contain 5% pyroxyline, 80% ether and 15% ethanol. It is advantageously applied in an amount making it possible to obtain a thickness of approximately 0.05 micron of the microencapsulation film.

This coating makes the beads biocompatible with blood and therefore usable for adsorbing the phosphates contained in the blood during an extracorporeal treatment.

The beads can be placed in cartridges which are inserted into the extracorporeal circulation circuit. They can also be mixed with other adsorbent materials.

The treatment can be carried out on whole blood or on plasma or hemofiltrate before their reinjection.

Les exemples suivants illustrent la présente invention. The following examples illustrate the present invention.

EXEMPLE 1

The beads thus formed are dried at a temperature of 800C and then heated to 350 C.

Porous beads are obtained which consist of zirconium oxide and carbon.

The pearls thus obtained are microencapsulated by mixing and stirring with 5% relative to the weight of the pearls of a collodion comprising by weight 80% ether, 15% ethanol and 5% pyroxyline (or fulmicoton)
As it evaporates, the ether and ethanol leave a thin film of 0.05 A of pyroxyline on the surface of the beads.
