Abstract

L'INVENTION CONCERNE UNE COMPOSITION DE COLORATION DES BACTERIES ET DES METHODES D'ANALYSE DES BACTERIES GRAM-NEGATIVES ET GRAM-POSITIVES. LA COMPOSITION CONTIENT UN AGENT CHELATANT ET UN COLORANT BASIQUE, TOUS LES DEUX ACTIFS A UN PH SUPERIEUR A 7,0. LA COLORATION DES BACTERIES PEUT SE FAIRE EN METTANT DES BACTERIES CONCENTREES OU EN SUSPENSION DANS UN FLUIDE, EN CONTACT AVEC LA COMPOSITION A UN PH NEUTRE OU BASIQUE. LES BACTERIES COLOREES AVEC LA COMPOSITION ET CONCENTREES PAR FILTRATION, CENTRIFUGATION OU UNE TECHNIQUE SIMILAIRE, SONT FACILEMENT VISIBLES ET LEUR PRESENCE DANS UN ECHANTILLON PEUT AINSI ETRE RAPIDEMENT DETECTEE. LES GRADATIONS DE COULEUR DES BACTERIES COLOREES, CONCENTREES, CORRESPONDENT AU NOMBRE DE BACTERIES ET UNE ANALYSE SEMI-QUANTITATIVE DES BACTERIES PEUT ETRE EFFECTUEE PAR COMPARAISON AVEC UN TEMOIN DE REFERENCE. LA DIFFERENCIATION DES BACTERIES GRAM-NEGATIVES ET GRAM-POSITIVES PEUT ETRE EFFECTUEE EN TRAITANT LES BACTERIES COLOREES PAR UNE SOLUTION D'UN ACIDE ORGANIQUE AYANT UN PH DE 2,5 A 2,6 ENVIRON. UN TEL LAVAGE DECOLORE COMPLETEMENT LES BACTERIES GRAM-POSITIVES. ENFIN, ON FOURNIT UN PROCEDE DE DETERMINATION DE LA SENSIBILITE DES BACTERIES VIS-A-VIS DES AGENTS ANTIMICROBIENS, QUI CONSISTE A INCUBER LES BACTERIES AVEC UN AGENT ANTIMICROBIEN, A COLORER LES BACTERIES, PUIS A COMPARER LA GRADATION DE COULEUR DES BACTERIES COLOREES, CONCENTREES, AVEC UN TEMOIN OU UN PRODUIT DE REFERENCE.THE INVENTION RELATES TO A STAINING COMPOSITION OF BACTERIA AND METHODS OF ANALYSIS OF GRAM-NEGATIVE AND GRAM-POSITIVE BACTERIA. THE COMPOSITION CONTAINS A CHELATING AGENT AND A BASIC COLORANT, BOTH ACTIVE AT A PH GREATER THAN 7.0. STAINING OF BACTERIA CAN BE ACHIEVED BY PLACING CONCENTRATED OR SUSPENDED BACTERIA IN A FLUID, IN CONTACT WITH THE COMPOSITION AT A NEUTRAL OR BASIC PH. BACTERIA COLORED WITH THE COMPOSITION AND CONCENTRATED BY FILTRATION, CENTRIFUGATION OR SIMILAR TECHNIQUE, ARE EASILY VISIBLE AND THEIR PRESENCE IN A SAMPLE CAN THUS BE QUICKLY DETECTED. THE COLOR GRADATIONS OF COLORED, CONCENTRATED BACTERIA CORRESPOND TO THE NUMBER OF BACTERIA AND A SEMI-QUANTITATIVE ANALYSIS OF THE BACTERIA CAN BE PERFORMED BY COMPARISON WITH A REFERENCE CONTROL. THE DIFFERENTIATION OF GRAM-NEGATIVE AND GRAM-POSITIVE BACTERIA CAN BE CARRIED OUT BY TREATING THE STAINED BACTERIA WITH AN ORGANIC ACID SOLUTION WITH A PH OF APPROXIMATELY 2.5 TO 2.6. SUCH A WASH COMPLETELY DISCOLORS GRAM-POSITIVE BACTERIA. FINALLY, A PROCESS FOR DETERMINING THE SENSITIVITY OF BACTERIA TO ANTIMICROBIAL AGENTS IS PROVIDED, WHICH CONSISTS OF INCUBATING THE BACTERIA WITH AN ANTIMICROBIAL AGENT, STAINING THE BACTERIA, THEN COMPARING THE COLORING OF BACTERIA, CONCENTRATE COLOR , WITH A WITNESS OR A REFERENCE PRODUCT.
