Abstract
Prepn. of a reagent for the diagnosis of Staph. aureus comprises (a) sensitising the red blood corpuscles of an animal with a serum contg. reactive immunoglobulins with respect to protein A and having an anti-red corpuscle activity for the said animal; (b) addn. of glutaraldehyde to the sensitised preparation and (c) lyophilisation of the preparation. Stable reagent having high sensitivity for the determination of Staph. aureus by haemagglutination. The diagnosis is very rapid about 30 seconds per test. Known reagents are unstable, diffult to prepare to give slow response times. The sensitisation in stage (a) is generally effected at 37-40 degrees C for 1-2 hrs. and stage (b) is effected at 37-40 degrees C for 10-15 hrs. after glutaraldehyde addition. Pref. the temp. is 40 degrees C, 0.0025-0.01 vol/vol.% of glutaraldehyde is used, the red blood corpuscles are sheep corpuscles and the serum used for the sensitisation is rabbit antisheep red blood corpuscle serum.
