Abstract

New cobalamin derivs. (I) are cpds. in which the OH gp. of hydroxocobalamin is blocked or replaced by the NH2 gp. of an amino acid, this NH2 gp. being sepd. from the COOH gp. of the amino acid by >2C atoms. Cpds. (I), which are hydroxocobalamin/amino acid complexes, are slow-release forms of hydroxocobalamin esp. useful for administration. Cpds. (I) have high hydrolytic stability in alkaline, neutral and even acid media; e.g., hydrolysis of the complex with 4-aminobutyric acid at pH 2 is complete after 3 days (compared with 36 hrs. for the complex with beta-alanine). This means that (I) can be administered orally without being hydrolysed in the stomach.
