Abstract

Peptides of formula (II) and their salts and intermediates (Va): where X=H, beta-Ala-NH, (alpha-methyl)-Ala-NH-, D-Ala-NH, D-Ala-Gly-NH-, beta-Ala-Gly-NH, Ala-D-Ala-NH. The alpha-amino gps. may be substd by an alkanoyl radical or (1-6C alkoxy)-carbonyl; V=Asp or Ala; W=Lys or Orn; Y=H, COOH, -CONH2, -CONHR or -CONR2; R=1-6C alkyl, 5-6C cycloalkyl or the two R gps. together form a tetramethylene or pentamethylene radical. (II) inhibit the secretion of pituitary growth hormone and to a lesser extent inhibit the secretion of other hormones e.g. glucagon; they have a similar spectrum of activity to the natural somatostatin. Since growth hormone and glucagon are insulin antagonists, (II) may be used in the treatment of diabetes and compared with natural somatostatin has prolonged duration of activity.
