Abstract

Multi-compartment containers containing a selection of culture media, detailed below, for the rapid identification of types of unknown bacteria. DETAILS OF MEDIA (1) Peptone 10g, K2HPO4 2g, glucose 10g, agar 17g, bromothymol blue 0.08g, distilled water to a litre. The green colour becomes yellow if glucose is fermented. Gas formation by enterobacteria fermentation is made evident by trapping of bubbles in the agar. Vosges-Proskauer positive organisms can be identified. Methyl red-positive organisms can be identified. (2) Meat extract 3g, tryptone 20g, ammoniacal FeSO4 0.2g, Na2S2O3 0.4g, cysteine 0.2g, agar 17g, distilled water to 1 litre. Production of H2S or indole can be shown. Browning of the medium indicates Proteus or Providencia. (3) Peptone 1g, glucose 1g, KH2PO4 6g, Na2HPO4 2g, tryptophan 3g, urea 20g, phenol red 0.01g, agar 17g, distilled water to 2 litre. For showing presence of urease and tryptophan deaminase. (4) Peptone 10g, meat extract 1g, lactose 10g, sucrose 10g, KNO3 1g, phenol red 0.04G, agar 17g, distilled water to 1 litre. Indicates fermentation of disaccharides and reduction of nitrate to nitrate. (5) Yeast extract 3g, L-lysine 5g, d-glucose 1g, KH2O4 0.4g, bromocresol purple 0.02g, agar 17g, distilled water to 1 litre. Shows presence of lysine decarboxylase and oxidase. (6) Yeast extract 0.1g, ornithine 5g, KH2O4 1.2g, Na2HPO4 0.4g, d-glucose 0.25g, bromocresol purple 0.02g, agar 17g, distilled water to 1 litre. Shows presence of ornithine decarboxylase. (7) NaCl 5g, MgSO4 0.2g, NH4.H2PO4 1g, K2HPO4 1g, NA citrate 5g, bromothymol blue 0.08g, agar 17g, water to 1 litre. Shows use of citrate. (8) Yeast extract 0.5g, di-glucose 0.25g, (NH4)2SO4 2g, K2HPO4 0.6g, KH2PO4 0.4g, NaCl 2g, Na malonate 3g, bromothymol blue 0.08g, agar 17g, water to 1 litre. Shows use of malonate. Organisms identified are:- Salmonella, Arizona, Citrobacter, Edwardsiella, Shigella, Escherichia, Klebsiella pneumoniae, K.ozenae, K.rhinoscleromatis, Enterobacter, Serratia, Proteus mirabilis, Pr.vulgaris, Pr.morganii, Pr.rettgeri, Providencia.
