Abstract
Covallatoxin derivs. of partial formula (I): (where R1 and R2 are each 1-5C alkyl, or CR1R2 is a is not 5 C alicyclic ring; R3 is H, lower alkyl, acyl, alkoxyalkyl, alkoxycarbonyl or aroyl, and R4 is formyl or opt. acylated CH2OH), which have the positive inotropic activity of convallatoxin with significantly higher oral resorption, are prepd. by reacting corresp. convallatoxin with R1R2CO in the presence of a dehydrating agent (pref. anhydrous CuSO4) and an acidic catalyst (pref. toluene-sulphonic acid), and opt. acylating or alkylating the resulting (I; R3 = H) and/or reducing the formyl gp. of (I; R4 = formyl) thus obtained.
