Abstract

Quinoline derivs (II) active against malaria and other blood parasites consist of (where R1 and R2 are halogen, OH, 1-4C alkoxy, 1-9C alkyl, phenethyl, styryl or benzyloxy and R1 can also be H), and their addition salts. They are prepared by (a) condensation of quinoline deriv. (III) with cpds. IV (H5C2)2N-(CH2)3-CY-CH3 (where Y is bifunctional groups such as -O- or geminal alkoxy groups) pref. in presence of acid catalysts, in melt or inert solvent, at 110-220 degrees C, followed by reduction of Schiff's base obtained, pref. with catalytically activated H2, at raised temp. and pressure, or with complex metal hydrides; or (b) by alkylating (III) with (H5C2)2N. (CH2)3. CHX. CH3 (where X is monofunctional group such as halogen or reactive ester group) pref. in inert solvent and presence of acid binders.
