Claims (5)

1. Menetelmä rekombinantteja nefrokystiinin (NPHP1] SH3-domeenin, jolla on SEQ ID NO:l:n sekvenssi, johdannaisia käsittävän kirjaston valmistamiseksi, mainitun menetelmän käsittäessä vaiheet: (a] tuotetaan SEQ ID NO:l:n mukaisen NPHP1 SH3-domeenin rekombinantteja johdannaisia vaihtamalla tai lisäämällä vähintään 1 aminohappo mainitun SH3-domeenin RT-luupissa, ja/tai vaihtamalla tai lisäämällä vähintään 1 aminohappo mainitun SH3-domeenin n-src-luupissa, ja (b] rakentamalla kirjasto, joka käsittää vaiheessa (a] tuotettuja SH3-domeenin rekombinantteja johdannaisia; jolloin NPHP1 SH3-domeenin rekombinantit johdannaiset säilyttävät vähintään 85 % identtisyyden SEQ ID NO:l:n aminohapposekvenssiin RT- ja n-src-luuppien ulkopuolella, ja jolloin RT-luuppi sijaitsee SEQ ID NO:l:n aminohappopositioissa 8-17, ja n-src-luuppi sijaitsee SEQ ID NO:l:n aminohappopositioissa 28-34.
2. Patenttivaatimuksen 1 mukainen menetelmä, jolloin vaiheessa (a] kuusi aminohappoa RT-luupissa on satunnaisesti vaihdettu, ja neljä aminohappoa n-src-luupissa on satunnaisesti vaihdettu ja kaksi satunnaista aminohappoa on lisätty n-src-luuppiin.
3. Kirjasto, joka on saatu patenttivaatimuksen 1 tai 2 mukaisella menetelmällä, mainitun kirjaston käsittäessä SEQ ID NO:l:n mukaisen nefrokystiini (NPHP1) SH3-domeenin rekombinantteja johdannaisia, käsittäen vähintään yhden aminohapon vaihdon tai lisäyksen mainitun SH3-domeenin RT-luupissa, ja/tai vähintään yhden aminohapon vaihdon tai lisäyksen mainitun SH3-domeenin n-src-luupissa, jolloin RT-luuppi sijaitsee SEQ ID NO:l:n aminohappopositioissa 8-17, ja n-src-luuppi sijaitsee SEQ ID NO:l:n aminohappopositioissa 28-34, ja vähintään 85 % identtisyyden SEQ ID NO:l:n aminohapposekvenssiin RT- ja n-src-luuppien ulkopuolella.
4. Rekombinanttien sitojaproteiinien valikointimenetelmä patenttivaatimuksen 3 mukaisesta kirjastosta, joka käsittää NPHP1 SH3-domeenin rekombinantteja johdannaisia, mainitun menetelmän käsittäessä vaiheet (a] saatetaan NPHP1 SH3-domeenin johdannaisia käsittävä kirjasto kontaktiin kohdemolekyylin kanssa riittävän ajan olosuhteissa, jotka sallivat johdannaisten ja kohdemolekyylin vuorovaikutuksen, jolloin kohdemolekyyli ei ole NPHP1 SH3-domeenin luonnollinen sitoutumisligandi, ja (b] valikoidaan kirjastosta yksi tai useampi nefrokystiini (NPHP1] SH3-domeenin johdannainen, jolla on spesifistä sitoutumisaktiviteettia kohdemolekyyliin, jolloin valitut NPHP1 SH3-domeenin johdannaiset säilyttävät vähintään 85 % identtisyyden aminohapposekvenssiin SEQ ID NO:l RT- ja n-src-luuppien ulkopuolella, ja jolloin RT-luuppi sijaitsee SEQ ID NO:l:n aminohappopositioissa 8-17, ja n-src-luuppi sijaitsee SEQ ID NO:l:n aminohappopositioissa 28-34.
5. Patenttivaatimuksen 4 mukainen menetelmä, jolloin mainittu kohdemolekyyli on proteiini tai peptidi.
